Advanced Magnesium Alloys Surface Modification by Ni-P Based Coatings

The dissertation thesis deals with the modification of the surface of advanced magnesium alloys with Ni-P based coatings. At the beginning of the theoretical part, the structures of the used magnesium alloys and the influence of individual alloying elements on their properties are characterized. In the following part of the thesis the current knowledge in the field of electroless deposition on metal substrates is summarized. The theoretical part of the thesis is closed with contemporary research study in the field of clarification and determination of possible mechanism of electroless deposition. For the subsequent investigation of the mechanism of electroless deposition on magnesium alloys, it was necessary to characterize the microstructure and composition of individual magnesium alloys in the first phase of the experimental part. The exact composition of elements was determined using glow discharge optical emission spectroscopy and scanning electron microscopy with EDS was used for composition of phases of magnesium alloys. Using scanning electron microscopy and detailed elemental analysis of the coated magnesium substrate, it was found that for optimal Ni-P coating deposition on magnesium alloys, acid pickling prior coating is required in a mixture of acetic acid and sodium nitrate. Using the XPS method, it was found that the phosphorus atom in the sodium dihydride-diphosphate reducing agent has a + V charge. 4 At the end of the experimental part scanning electron microscopy and detailed elemental analyses were used for monitoring of the Ni-P particles nucleation and growth in the first 120 seconds of the coating process.
Friction Modification within Wheel-Rail Contact

Předložená disertační práce se zabývá experimentálním studiem modifikátorů tření a maziv pro temeno kolejnice, které jsou aplikovány do kontaktu kola a kolejnice za účelem optimalizace adheze a redukce hluku. Hlavním cílem práce bylo objasnit vliv aplikovaného množství a složení těchto látek na adhezi v kontaktu. Hlavní pozornost byla věnována zejména potencionálním hrozbám souvisejících s kriticky nízkou adhezí, která může nastat po aplikaci těchto látek. Experimentální studium probíhalo v laboratorních i reálných podmínkách, konkrétně v tramvajovém provozu. V případě laboratorních experimentů byl využit komerční tribometr a dvoudiskové zařízení umožňující simulovat průjezd vozidla traťovým obloukem. Kromě samotné adheze bylo při experimentech sledováno také opotřebení a míra hluku. Výsledky ukázaly, že maziva pro temeno kolejnice jsou schopna poskytovat požadované třecí vlastnosti, nicméně jejich chování je silně závislé na aplikovaném množství. V případě předávkování kontaktu dochází ke kriticky nízkým hodnotám adheze, které vedou k výraznému prodloužení brzdné dráhy. V případě modifikátorů tření bylo ukázáno, že chování těchto látek je výrazně ovlivněno odpařováním základního média. Výsledky také ukázaly, že nadměrné množství částic pro modifikaci tření může způsobit kriticky nízké hodnoty adheze. U obou výše zmíněných typů produktů byl prokázán pozitivní vliv na míru opotřebení a míru poškození povrchu, zatímco významná redukce hluku byla dosažena pouze v případech, kdy došlo ke značnému poklesu adheze. V závěru této práce jsou uvedena doporučení pro další výzkumné aktivity v této oblasti.
Metal Matrix Composites Prepared by Powder Metallurgy Route

Ve všeobecnosti, poznatky o design slitin, jejich výrobě a výběru legujúcich prvků sú omezené na slitiny s jedním základním prvkem. Tento fakt ale výrazně limituje možnosti a volnost výběru prvků pro dosáhnuti speciálních vlastností a mikrostruktur. V poslední dekádě se ukázalo, že materiálová věda a inženýrství nejsou ještě zdaleka prozkoumané v důsledku objevu nové třídy materiálů nazvané vysoko entropické slitiny (HEA high entropy alloys). Jejich objev upoutal pozornost vědecké komunity. Základní koncept pro jejich design je, že namísto jednoho, nebo dvou základních prvků obsahují minimálně 5 prvků v podobných atomových koncentracích. V posledních letech se objevila skupina materiálů odvozená od HEA, nazvaná slitiny so střednou entropii (MEA medium entropy alloys). Na rozdíl od HEA ale obsahují 3, nebo 4 prvky. Táto práce je věnovaná studiu přípravy a charakterizaci HEA, MEA a jejich kompozitů s pomocí metod práškové metalurgie. V této práci byli dohromady zkoumány tři kompozice: AlCoCrFeNiTi0.5, Co1.5Ni1.5CrFeTi0.5 a CoCrNi, kompozity s kovovou matricí (MMC metal matrix composites) vyztužené částicemi B4C s CoCrNi jako matricí. Hloubková mikrostrukturní a mechanická analýza těchto materiálů byla provedena pomoví metod rastrovací a transmisní elektronové mikroskopie spojené s tahovými a ohybovými zkouškami. V průběhu celé studie se objevovaly problémy s kontaminací kyslíkem, co se projevilo vznikem značného množství oxidů v připravených materiálech. U Slitiny AlCoCrFeNiTi0.5 byla naměřena tvrdost přesahující 800 HV. Její houževnatost ale byla velice omezena. V její mikrostruktuře byly identifikovány částice in-situ TiC v důsledku přítomnosti organického, anti-aglomeračního činidla (metanolu) v mlecí misce. Tato reakce může být použita v budoucnu k přípravě MMC se záměrnou disperzí TiC. Na druhé straně, slitina CoCrNi ukázala vysoké hodnoty tažnosti (26%) a meze pevnosti přes 1000 MPa. Mikrostruktura obsahovala majoritní FCC fázi s BCC precipitáty. Tahle slitina byla z důvodu vysoké tažnosti zvolena pro přípravu kompozitu s výztuží B4C. V průběhu slinování ale došlo k reakci mezi přítomným Cr a B4C, které výsledkem byl Cr5B3 borid. Tento kompozit mel pevnost v tahu 1400 MP a extrémne jemnozrnnou strukturu. Celková tažnost ale klesla na 1.9 %. Slitina AlCoCrFeNiTi0.5, která mela strukturu složenou jen z FCC tuhého roztoku dosáhla nejlepší kombinaci mechanických vlastností s pevností přesahující 1300 MPa a dostatečnou tažností 4%. Prášková metalurgie se ukázala jako vhodná metoda pro přípravu HEA a MEA slitin a jejich kompozitů, s dobrou kombinací pevnosti a tažnosti. Tato metoda dovoluje měnit mikrostrukturní parametry připravených materiálů jednoduchou úpravou parametrů procesu.
Regulation of stomatal development by environmental conditions and physiological processes in the leaf

Stomatal development and its regulation by environmental conditions (light, CO2 concentration) and physiological processes in the leaf of higher plants were investigated. The study was based on the assumptions that stomatal development should be regulated by signals coming from both external environment and leaf interior, and that the signal should be transduced from cotyledons to leaves. Transgenerational effect in stomatal development was also studied. Molecular and physiological approaches were applied to reveal the relationship between leaf environment, stomatal development, stomatal function and leaf physiology.
Magnetic biocomposite materials for removal of significant xenobiotics from water systems

The theoretical part of this doctoral thesis provides a comprehensive overview on the topic of preparation and subsequent utilization of magnetic derivatives of biological materials for xenobiotic separation from water. Main attention is paid to magnetic modification of waste materials and by-products originating from agricultural and food industry, which represent widely available and low-cost materials, and also to magnetic modification of microbial cells. In addition to the description of magnetic particle preparation and individual developed techniques of magnetic modification, a brief characterization of selected pollutants and a detailed table overview on utilization of magnetically responsive biomaterials for biosorption of organic dyes, heavy metals, pharmaceutical and personal care products together with ubiquitous industrial endocrine disruptors and also of crude oil derivatives is presented. Experimental part of this thesis is focused on the preparation and optimization of new types of magnetic materials. Emphasis is placed on the employment of simple, fast and simultaneously low-cost magnetic modification techniques (e.g., postmagnetization using microwave-synthesized magnetic iron oxides or one-step modification by magnetic fluids). Selected plant materials (barley and rye straw) were chemically modified to significantly (up to five-times) increase the maximum adsorption capacities for tested dyes. All prepared biomaterials exhibited a great magnetic response and simultaneously relatively high adsorption capacity for selected xenobiotics under experimental conditions used. Factors substantially affecting adsorption process, such as pH, initial concentration, incubation time or temperature were also studied. Adsorption equilibrium data were assessed using Langmuir, Freundlich or Sips isotherm models. Experimental data from time dependence study were analyzed by chosen kinetic models, namely the pseudo-first-order and pseudo-second-order ones and by intraparticle diffusion model. Thermodynamic parameters (Gibbs free energy, enthalpy and entropy) describing the nature of adsorption were also included in study.
Vliv genotypů pro gen leptin na vybrané kvalitní ukazatele hovězího masa

The dissertation "The Influence of Genotype of Leptin Gene on Qualitative Characteristics of Beef" describes the effect of polymorphisms of gene of leptin on selected qualitative, morphometric and weight indicators of beef and carcass. The analysis was carried out on 333 bulls of Czech Fleckvieh, which were genotyped (by PCR/RFLP method) of chosen locus. This locus is associated with qualitative changes in beef according to previous foreign studies. The weight of carcass, fore and hind quarter of the right half of the carcass, the weight of the individual tissues (meat, bone, and fat) and the weight of main meaty parts from the right half of carcass were measured. Before the dissection chosen morphometric indicators were measured. After the dissection, a sample of the musculus longissimus lumborum et thoracis was taken and then analysed in the laboratory. Basic chemical composition (dry matter, fat content, protein, and ash), muscle fibers diameter and fatty acid profile was determined. In addition, pH, added water holding capacity, colour (CIE Lab colour coordinates) were analysed both for one-day post-mortem and after 14 days of ageing. The shear force (using the Warner-Bratzler probe) was evaluated at the same time intervals as for both raw and heat-treated samples by grilling. The studied LEP polymorphism had a statistically significant effect on the weight of the hot carcass, the weight of right half of carcass and fore and hind quarter. LEP polymorphism significantly affected the proportion of meat on the right half of carcass, the weight of the bones and total weight of the I. class meat, and the weight of the I. class meat in the fore quarter. Further, the effect of LEP polymorphism on the weight of boneless round, boneless chuck, plate, flank, rib, hind shank, and meat trimmings of fore quarter was found. In a group of morphometric indicators, the effect was significant on lengths of round, fullness of round and round circumference. In the case of chemical composition, the effect of LEP polymorphism on the intramuscular fat content was significant. From the qualitative indicators, the effect was significant on the pH value and added water holding capacity, both one-day post-mortem and after 14 days of ageing. In the case of tenderness, the effect on shear force of heat treated, non-aged meat was significant. From the indicators describing colour, a statistically significant effect on the indicator of lightness was found in the case of unconditioned meat and after 14 days of ageing the effect was significant on the indicator characterising yellow colour and the colour saturation. Determination of fatty acid profile was carried out by gas chromatography. The effects of studied polymorphism and differences between LEP genotypes were found in myristic, palmitic, palmitoleic, linoleic, -linolenic, -linolenic, conjugated linoleic, eisocapentaenoic, docosatetraenoic, docosapentaenoic both n-6 and n-3 and docosahexaenoic acids. From aggregated indicators, the statistical significant effect of LEP polymorphims on PUFA content, omega-3 fatty acids content, and omega-6/omega-3 fatty acids ratio was found. In connection with other candidate gene loci influencing the quality of beef, the results of dissertation can be used in the preparation of breeding schemes aimed at the production of quality breeding animals, highlighting the higher quality standards of the produced meat.

The main goal of this dissertation thesis has been defined as the development and presentation of a model designed to show transparency of different effects of tourism in urban destinations and its practical application by destination management organizations. The resident sub-model was compiled, based on research in the selected destination as well as its statistical evaluation. This sub-model clearly demonstrates relationships that affect tourism. This decision model was focused on the socio-economic effects, primarily generated by residents, which influence tourists and basically tourism in urban destinations. The resident sub-model was subsequently incorporated into another decision model of the sustainable destination to show the resident influence on objective economic factors generated by tourism in urban destinations. These two models clearly illustrate how complicated system is the behaviour of residents and which significant negative / positive effects on tourism can generate. For the development of this solution system thinking was used as the main methodological approach. System thinking has been actually rarely used, but it is very effective method, because it can reveal complexity of diverse relationships and find possibilities how to influence and control these relationships; in this case by destination management organizations. This solution is intended primarily for these tourism institutions. This method was intentionally simplified to be applicable by the local organizations. However, it retains high level of efficiency, which can bring required objectivity in very subjective relationships between residents, tourist and basically tourism at urban destinations.
