The examples of introduction of birds of family Falconidae in the urban environment

The present thesis focuses on the issue of reintroduction of the Peregrine Falcon (Falco peregrinus) to different locations and types of habitat. Four methods of reintroduction of birds of prey to their natural habitats are being used: they might be released from a release cage; they might be included in a falconry training programme; or they might be adopted, either within their own species, or cross-species. Of the methods mentioned, releasing from the release cage is the most frequent in the Czech Republic. The young birds of prey to be released should grow in an environment similar to the one they are about to be released. Each of them has to pass a veterinary examination and should be properly labelled and registered with the Regional Authority. It is crucial for the birds to minimise their attachment to and dependence on man. Many of the reintroduced birds successfully reached sexual maturity and were able to mate and breed. According to the research, the survival rate of 1-year-old falcons is 65 per cent in urban and 28 per cent in rural areas. In the present thesis, I have included 11 Peregrine Falcons and 7 Saker Falcons (Falco cherrug) that have been reintroduced on the territory of the Czech Republic. The findings of this thesis may contribute to further research.
The potential use of artificial nests for the management of Peregrine Falcon in industrial areas

In my Bachelor Thesis I have focused on presentation of a project concerning location of artificial birdhouses at two particularly selected chimneys situated within an industrial park called Chempark Záluží in Litvínov. Those two birdhouses were installed in the industrial park, as peregrines (falcons) settling in these nests was anticipated. The project launched in 2011 and successfully continues up to now. This project is one of most successful projects focused on creating new nesting opportunities for falcons, fixed on one specific industrial park. Fifteen falcon pups were successfully brought up to 2015. Presentation of the project aims to spread such an activity on other suitable places and integration of wide public into other activities, directed on protection of falcons on our territory, which are as well presented in this thesis, including assessment of their importance.

The main aim of this thesis was to evaluate the genetic variability in different populations of birds of prey in the Czech Republic. As an alternative source, the feathers were used for the extraction of DNA. The reference species were chosen the Peregrine Falcon (Falco peregrinus) and the Saker Falcon (Falco cherrug), which was also compared with the subpopulation living in Slovakia. Ten microsatellite markers from the literature were tested. Polymorphism of markers varied significantly, locus NVH fp5 was the least polymorphic (PIC = 0.185 F. p; PIC = 0.119 F. ch.). Null alleles were observed in this locus in the Peregrine Falcon population, that is why it should be discarded from the microsatellite panel. The genetic diversity was low among subpopulations of the Peregrine Falcon. FST for the population living in the captivity and living in the wild was 0.025. In the case of the Saker Falcon subpopulation living in Slovakia showed moderate genetic diversity values (0.185 for the subpopulation living in the captivity and 0.126 for the subpopulation living in the wild in the Czech Republic). All subpopulations (except museum specimens of the Saker Falcon) were in Hardy - Weinberg equilibrium. Gene flow was higher among subpopulations of the Peregrine Falcon than the Saker Falcon, where the other population from the different geographical area was also included. 454 sequencing revealed 3 complete mitochondrial DNA sequences of Peregrine Falcons, 2 of Saker Falcons and 2 of Gyrfalcons (Falco rusticolus). The longest sizes were 16,154 bp for the Saker Falcon, 17,239 bp for Gyrfalcon and 17,527 bp for the Peregrine Falcon. Sequence of the Peregrine Falcon was inserted into the Genbank database under accession number JX029991. Whole genome mitochondrial DNA sequences of Saker Falcons and Gyrfalcons have never been published, that is why it will be the part of the new manuscript.
Určování příbuzenských vztahů u druhů CITES na základě analýzy mikrosatelitů

Cílem projektu bylo vypracování metod DNA analýzy využívající mikrosatelitových markerů u vybraných druhů chráněných dle CITES a zavedení metodického postupu pro určování rodičovství do praxe. Metoda byla ověřena na druzích raroh velký, sokol stěhovavý, sokol lovecký, jestřáb lesní. Byl vypracován a ověřen metodický postup s využitím 12-ti mikrosatelitových lokusů pro určování rodičovství.

Cílem projektu bylo vypracování metod DNA analýzy využívající mikrosatelitových markerů u vybraných druhů chráněných dle CITES. Zvoleny byly druhy: raroh velký, sokol stěhovavý, sokol lovecký, jestřáb lesní. Uvažovaný projekt má za cíl uvést do praxe soubor metodických postupů na určování rodičovství pomocí analýzy DNA k terénnímu použití pro státní orgány ochrany přírody v oblasti upravené zákonem č.16/1997 Sb.
