Design and Operation of Water Disinfections Plants

In the first part of my bachelor thesis I deal with individual disinfectants used for hygienic supply of drinking water based on chlorine and without chlorine. Then I mention the operation without chlorine disinfectants. In the second part of this thesis I describe individual plants using various disinfection methods and at the end of the thesis I compare these methods and use it in practice.
Role of ventral hippocampus and medial prefrontal cortex in behavioral flexibility in rodents

Behavioral adaptation to a continuously changing environment is critical for the survival of the animals, but also day-to-day interactions in the human world. The main components maintaining flexibility in cognition and behavior are well-established and depend mostly on proper intercommunication within the prefrontal cortex (PFC) and striatum. Some parts of the PFC are densely innervated by the ventral hippocampus (vHPC), which has a great impact on its functioning. Also, hippocampal-prefrontal circuit dysfunction has been shown to disrupt the integrity of flexible cognition in some neuropsychiatric diseases. Therefore, the exact functional role of this pathway is an indispensable part of the research. The aim of this study was to test the role of the vHPC and the medial PFC (mPFC) in an active place avoidance task on a rotating arena in 1) two flexibility task variants - reversal learning and set- shifting - and 2) the spatial memory retrieval. We inactivated these structures by muscimol (GABAA receptor agonist) in a variety of unilateral, bilateral, and combined local injections. Disrupted performance was apparent in reversal learning in vHPC-inactivated rats. No effect was seen in mPFC-inactivated rats. Impairments after the task acquisition were observed in bilateral vHPC and mPFC inactivations...

Role slizniční imunity a střevní mikroflóry při vývoji zánětlivých onemocnění

Gut microbiota is important for our health and well-being, but when its composition is disrupted, it can induce or perpetuate several chronic inflammatory disorders, including inflammatory bowel diseases (IBD). The mechanisms which distinguish protective microbes from the deleterious or indifferent ones are largely unknown. The aim of this thesis was to study the interaction of the immune system with microbes that have different relationships to IBD pathogenesis. Escherichia coli is a predominant aerobic microorganism of the gastrointestinal tract. This species includes microbes implicated in induction of IBD as well as in its therapy. Four E. coli strains with different relations to IBD were selected for our experiments: E. coli Nissle 1917 (EcN), which has been successfully used in IBD therapy, E. coli strains LF82 and p19A, which have been implicated in the pathogenesis of IBD, and E. coli strain K6, which has neither been implicated in pathogenesis nor in protection from this disease. The experiments were performed both with living bacteria and inactivated ones. As the mode of inactivation may change the microbial antigenic structure, we measured how different methods of inactivation, i.e. 1% formaldehyde, exposure to heat or UV irradiation, influence the microbe's immunogenicity.
