Detekce variability DNA ve vybrané kolekci ostropestřce mariánského (Silybum marianum L.)

Milk thistle (Silybum marianum L.) is a promising crop with a wide range of practical applications. It is the only source of the flavonolignan silymarin, which has a positive impact on the health of both humans and animals. Although it is particularly significant as a medicinal plant due to its beneficial effect on liver parenchyma, its genetic diversity has not been thoroughly explored. The study of variability is fundamental for optimizing agricultural production of this crop, clarifying correlations between genome architecture and flavonolignan content, breeding, or categorizing gene sources. This study evaluates the variability of a selected collection of milk thistle using iPBS primers and sequencing analysis of trnC-F and psbA-trnH regions in cpDNA. Knowledge of these sequences can be further utilized, for example, in phylogenetic analyses, diversity studies, and hybridization methods. The study of genetic variability at the molecular level provides important insights for the ideal cultivation or breeding of milk thistle.
Use of interspecific ICSI method for assessing the quality of the retained biological material from genetic resources

Intracytoplasmic sperm injection, or ICSI, is a commonly used method in assisted reproduction centers and is also an effective tool for a fertilization research. The interspecific ICSI is the suitable method for production of zygotes, which are formed by association of gametes of different, distantly unrelated species. It can bypass natural interspecies barriers and create so-called interspecific zygotes. This can be used to evaluation properties of genetic material. In this work, this method was used for evaluation of the quality of stored sperm originating from male goats of the white shorthair goat genetic resource. The aim of this work was to evaluate the effect of cryopreservation on the quality of goat sperm, which was frozen in three different ways at two different breeding stations. Freshly collected semen served as a control group. The heads of xenogeneic goat sperm were injected into mature mouse oocytes. The injected oocytes were allowed to develop to the zygote stage, in which both female (mouse) and male (goat) pronuclei was formed. In male pronuclei, DNA damage was assessed by immunofluorescence staining with anti-γH2AX antibody, a marker of DNA breaks. The object of this thesis is to describe genetic diversity of pigs, evaluation of genetic diversity and description of current state of genetic resources preservation. Firstly is described importance of pig-breeding, domestication of pigs and breeds of pig mostly kept in the Czech Republic. Further is described biodiversity and dividing of biodiversity, genetic diversity and research methods of genetic diversity. Last chapter is about genetic resources.

The aim of the diploma thesis was to compare performance of breed Czech Golden Spotted Hen originating from different breeders, which were housed both in MTD Ústrašice and in sub-optimal conditions in backyards. The aim was also to evaluate differences in egg quality (egg index, egg weight, weight, proportion and color of yolk, weight, proportion, color and strength of eggshell and Haugh units) among the breeders. Individual laying intensity was observed too. The last part of the thesis compares growing intensity in rearing period in Czech Golden Spotted Hen, Polish Green Feet Hen and hybrids Dominant in sub-optimal condition.
Yeald formation of emmer in organic farming

In this thesis analyzes the problem of possible uses and cultivation of emmer and its suitability for cultivation in organic farming. It is solved by creating revenue emmer just in terms of organic farming. It has also been solved by making a survey of varieties and varieties of emmer across Europe. In this work are summarized practical knowledge gained from field experiments were conducted with wheat genetic resources in terms of organic farming on habitats in the Czech Budejovice in South Bohemia area. All varieties used in an attempt to come from the Gene Bank in VÚRV in Prague-Ruzyne. The experiments were varieties of wheat and emmer wheat.
Structure of genetics resources and the actual use of einkorn (\kur{Triticum monococum }L.) in the Czech Republic

Purpose of this thesis is analysis of the whet genetic resources (Triticum monococcum L.), availability of einkorn seeds in the Czech Republic, the condition of cultivation and treat in the Czech Republic. The first part of my thesis deals with an organic farming in our country and also in the European Union. The following chapter are involved in choice of species and varieties and the information about non-traditional cereals. The accent is on characteristic, agro-technology and utilization of einkorn. For the presentation of results are used the data from database EVIGEZ.
