The aim of this diploma thesis is determination of alfalfa (Medicago sativa) infestation in Agricaltural company Kvasicko a. s. The monitoring of infestation was doing from 2013 to 2016 on land Odšenkovna, sawn on 29. 8. 2013, and on land Novina levá, sawn on 11. 4. 2014. Evaluation was made by counting method. The results of this evaluation were processed with DCA analysis and canonical corespondence analysis (CCA). It was found 11 species of weeds on observed lands during the monitoring. The most frequently weeds were: Capsella bursa-pastoris, Stellaria media, Taraxacum sec. Ruderalia and Veronica.
Germination of long-term storaged seeds of selected grass and legumes

Quality reproductive material is created in the fields but its subsequent quality and the general ability to germinate may be affected by many other factors. The fundamental and necessary capability of the seeds is their germinability. In my thesis I selected seeds approximately 6 - 8 years old of certain types of grass or clover plants (Trifolium repens, Trifolium pratense, Medicago sativa - Zuzana, Medicago sativa - Pálava, Medicago lupulina, Trifolium hybridum - Oderský, Anthyllis vulneraria and Onobrychis viciifolia) and I monitored their germinability, its dynamics and capacity. Subsequently, I also used fresh seeds to compare the results and to demonstrate how seeds of different ages may differ. The experiment was performed under home conditions. All seeds to be germinated were placed on Petri dishes on which I maintained stable humidity. I monitored germinability of the seeds in previously set intervals after 2, 3, 5, 7, 10 and 15 days. Originally, the plan was to measure germinability also after 21 days but the germinating material did not last that long and losses were detected as early as after 15 days. I saw big differences between the individual species. Some of them germinated very well and their dynamics was high (Trifolium repens, Medicago sativa - Zuzana, Medicago sativa - Pálava), while others did not germinate at all or very little (Onobrychis viciifolia, Anthyllis vulneraria). In some species I even observed differences in germination dynamics.
Identification and sequencing genom of a new virus infecting lucerne

Samples of lucerne plants characteristic with local necrosic lesions, leave malformation and yellow spots on leaves were investigated with transmission electron microscopy. Virus particles observed there were filamentous ones of 600 to 700 nm long. Nucleic acid was isolated, transcribed and amplified using PCR. Genus-specific primers were designed based on reverse genetics from the highly conserved genes for carlaviruses, potexviruses and potyviruses. Successful amplification with carlavirus-specific primers, sequencing and comparison with sequences in GenBank database revealed presence of a carlavirus. This was later identified by nucleotide sequence comparison as a new isolate V4 of Alfalfa latent virus. Specific primers for isolate V4 were designed in a coat protein position. Half of the genom of this virus was obtained with PCR and PCR modified amplifications and compared with sequences of Alfalfa latent virus and Pea streak virus from GenBank.
Comparison of drought and frost resistence of selected varieties of \kur{Medicago sativa} L. from domestic and foreign breeding

The aim was comparison of drought and frost resistence of selected varieties of Medicago sativa L. from domestic and foreign. The evaluation was performed by a series of laboratory experiments and observations in two replications in 2009 ? 2010. The results were evaluated as the most resistant varieties to frost variety from the Czech Republic and Slovakia. Namely variety Magda Litava, Tisa and Synthesis. As the least resistant varieties have been evaluated varieties from the warm coastal areas. Most resistant varieties to water deficit during vegetation varieties were evaluated Magda, Comete, Europe and Camilla. The climatic conditions of the Czech Republic would be most appropriate, according to the results observed varieties of Magda. It has high resistance to frost and has good results in drought. Other good varieties from Czech Republic are Pálava and Camilla. Of the foreign varieties would be best to use a French variety of Como, Europe and the Dutch variety of Capri. These varieties have good resistance to frost and high drought. The whole problem of drought and frost resistance of alfalfa is not yet explored, it would be appropriate, the results obtained confirmed by further research.
