Application of molecular and cellular biology methods in research of protozoa Eimeria

Eimeria is an apicomplexan parasite causing disease coccidiosis that is most prominent in poultry farming industry. This thesis is aimed to develop new molecular tools and resolve issues that would be a valuable contribution in the field from both research and industry perspective. Because immunity to Eimeria is strictly species- specific, it is important to know and recognize correctly all species that parasitize the host. Traditional diagnostic approaches rely on classical methods such as oocyst morphology determination under the microscope, measurement of prepatent period or in-vivo assessment of lesions caused by this parasite. However, diagnostics of individual species using these methods is very time-consuming and it is often unreliable, especially when mixture of multiple species whose parameters overlap is analyzed. Methods utilizing conventional PCR to distinguish species already exist, however, they lack advantages offered by quantitative real-time PCR (qPCR). The first aim of this thesis was to develop qPCR assays for detection and quantification of seven Eimeria species which infect chicken utilizing single-copy non-polymorphic targets in order to ensure maximal specifity and coverage of all strains of each species. The evaluation of meat performance of turkeys

The goal of thesis was to create a literature review that will evaluate the influence of genotype, sex and age on growth, carcass yield and composition of meat turkeys. Genotype is one of the most important factors for growth, which is reflected already in the prenatal stage and then continues in the postnatal stage. And at the age of 20 weeks, differencis between males and famale are 35% in temrs of males. Sex plays important, role brouse male turkeys have a significantly higher growth rate. Changing the speed growth also depends on age. Carcass yield moves in broiler turkeys between 76-83%, genotype affects carcass yield and greatly affects the weight of the breast and thighs. Sex determinate primarily weight of valuable parts. Carcase composition varies depending on age. Turkey meat is considered to be good quality with a high content of nutrients. Turkey breast contains 22.6% protein and only 1.7 fat. Effect of genotype on the composition of turkey meat is in the representation of protein and fat. Protein and fat content also depends on sex. Meat composition varies depending on age.

This thesis deals with a factors, that affect the final performance of turkeys for fattening of meat yield. It takes into account internal and external aspects based on essential needs of the modern commercial hybrids from well-known companies. The thesis brings a familiarization with origin and exterier of turkeys and mentions an essential breeds and types of turkeys. It includes a breeding and situation of turkey breeds in the Czech Republic. My thesis deals with a compare of general informations with practical experience and finding out that maximal performance of turkey connected with technological principles of fattening.
