Characterization of viral nanoparticles derived from mouse papillomavirus

The L1 and L2 capsid proteins of papillomaviruses are characterized by the ability to self- assemble into viral capsids, which can be divided into pseudovirions (PsVs) and virus-like particles (VLPs) by inner content. In addition to the fact that such particles can serve as "nano-containers" for diagnostic and therapeutic agents, it has also been shown that papillomaviruses, whether wild, PsVs or VLPs have a higher affinity for tumor tissue than non-tumor tissue. This thesis deals with relatively newly discovered (2011) mouse papillomavirus (MusPV) and nanoparticles derived from this virus. This papillomavirus has been chosen for its positives, including easy preparation of VLPs and PsVs, as well as an available model organism for possible testing. Furthermore, MusPV has the potential for use in gene therapy and cancer diagnosis, because there is no immune response in the human population. The aim of this diploma thesis is to prepare an expression system for the production of PsVs and VLPs. In additional it will also look at the quality and quantity of PsVs and VLPs, characterization of these particles and verification of existing postulates regarding higher affinity of papillomaviruses for tumor cells. Effect of pH on initiation of viral infection

In organisms, acidic pH occurs either in the intracellular or extracellular environment. Intracellularly an acidic pH is found in particular cellular compartments, such as early and late endosomes or lysosomes. In the extracellular space, the acidic pH is found in hypoxic tissues, or in tissues where inflammation or tumour growth occurs. The value of the pH is usually in the range of approximately from 5.5 up to 6.5. This work focuses on the description of the conformation changes of viral proteins, which occur due to a decreased pH and lead to escape of viral components from membrane compartments. Changes in pH in organelles of endocytic pathways as well as changes in extracellular pH affect significantly the infection of many viruses. Research on the role of pH during the viral can bring important information for the development of antiviral and anticancer treatment. Key words: virus, conformation, pH, receptor, endocytosis
Application of method Competitive Intelligence on the building industry

The main theme of the thesis is the application of methods of information and knowledge-based disciplines - Competitive Intelligence on the building industry of the Czech Republic. The first part provides a comprehensive overview of theoretical information which are necessary to describe Competitive Intelligence. From the outset, it gradually forms detailed description about the entire field as far as detailed procedures with the application of various analytical methods, in the form of formalized definitions of Competitive Intelligence. The most common methods which are listed below in particular are SLEPT analysis, SWOT analysis, an analysis of five porter forces, BCG matrix, and others. The practical part of work is an application of process and methods of Competitive Intelligence on the Czech building market. This part gradually specifies individual steps of analysis (definition of objectives, procedures, identification and exploitation of information resources, etc.) and presents its final output, which is suitably complemented with a description of information resources. Chapter concludes the summary of the main trends and factors involved in the building industry, which complements the recommendations for the building companies to increase their competitiveness. There is also important summary and characteristics of selected information sources.
Vývojové prostření Eclipse

Tato práce popisuje především praktickou stránku vývojového prostředí Eclipse. Cílem bylo vytvoření příručky, která by programátorům začínajícím s Eclipse usnadnila proniknutí do jeho praktického používání pro programování v jazyce Java. Z tohoto důvodu je určitá znalost jazyka Java nutným předpokladem. První kapitola práce je spíše teoretická a stručně popisuje nejvýznamnější charakteristické rysy Eclipse. Druhá část se věnuje nejdůležitějším praktickým operacím a úkonům, které jsou běžně používány při programování v jazyce Java. Třetí část pak popisuje možnost rozšíření základního programovacího prostředí Elipse pomocí pluginů a praktické používání některých z nich.
