Mitochondrial uncoupling and mitochondrial morphology in relation to type-2 diabetes

- 2 - 1 SUMMARY Increased ATP/ADP ratio resulting from enhanced glycolysis and oxidative phosphorylation represents a plausible mechanism controlling the glucose-stimulated insulin secretion (GSIS) in pancreatic -cells. Using high resolution respirometry and parallel mitochondrial membrane potential ( m) monitoring we have quantified bioenergetics in rat insulinoma INS-1E cells representing a suitable model for studies of insulin secretion in vitro. Upon glucose addition to glucose-depleted cells, we have demonstrated a simultaneous increase in respiration and m during GSIS and shown that the endogenous state 3/state 4 respiratory ratio hyperbolically increases with glucose, approaching the maximum oxidative phosphorylation rate at maximum GSIS. Attempting to assess the basis of the "toxic" effect of fatty acids on insulin secretion, GSIS has been studied after linoleic acid addition. The linoleic acid addition diminished the observed respiration increase, m jump, and magnitude of insulin release, and reduced state 3/state 4 dependencies on glucose, which was caused by mitochondrial uncoupling. Derivatives in family finances

This thesis focuses on the derivatives market. The goal is to choose ones that are suitable for use for the benefit of family finances and planning. Mapping the world market derivatives, and then describes the basic characteristics of the main types of contracts. After the general characteristics of the derivatives in the introduction, are briefly described two types (forwads and swaps) that are not directly used in the family finances. In their description, however, are pointed out the fundamental principles of derivatives and trading with them. In the next section, the focus is on those derivatives that are suitable for use in household budgets. They include Futures, Options, Warrants, Investment Certificates, Exchange Traded Funds (ETF), Contracts for diference (CFD). There are also discussed basic principles of these instruments, their use and the current bid.
Využití vysokorozlišovací cytometrie ke studiu živých buněk

The fluorescent proteins are important innovation in the field of cell biology and in vivo experiments that use fluorescent proteins bring new interesting results from a different point of view than in vitro and in situ experiments. This presentation will introduce our automated 2D/3D high-resolution time-lapse image acquisition and analysis of living cells transfected with plasmids encoding fusion proteins or dyed with fluorescence probes. One of our in vivo projects, we will mention, is focused on the study of mitochondrial and nuclear apoptotic processes in living cells. Another in vivo project concentrates on the study of topography of telomeres and incidence of telomere-association phenomenon in various types of human tumor and healthy cells. In our experiments, cells are stably or transiently transfected with plasmid DNAs coding mitochondrial apoptogenic or telomere-binding proteins with fluorescent proteins.
