Renewal of a social house in Zlobice
Váchová, Barbora ; Muroň, Ivo (referee) ; Ležatka, Lukáš (advisor) ; Struhala, Karel (advisor)
The aim of the given thesis is to describe the reconstruction of a hall in Zlobice as well as a foundation of the new administrative area. The hall is not a monument, yet the authors consider it to be one. Nowadays the hall is not used and it decays, therefore the project focuses on a possible technical renovation as well as its future use. The design considers the possible use of the building for social gatherings, cultural events and flats at the same time. Next there is an L shape extension that together with the historical building creates a courtyard. The extension will serve for the administrative purposes and the building maintenance. The next part of the project deals with a renovation of the picnic area that severs for free time activities and social gatherings. The altar situated in the area is designed to serve for possible performances and a background for performers. The aim of the project is to describe the renovation of the hall and its surroundings and at the same time it proposes a new possible usage of these premises.
Sport hall and its facilities for the elementary school Hudcova 35, Brno
Váchová, Barbora ; Klimecký, Martin (referee) ; Gerö, Jiří (advisor)
The proposal is based on a pre-diploma project, where the Medlánek area was comprehensively addressed and a housing development, a kindergarten, a church and civic amenities were designed for the area. The sports hall, which is the subject of the thesis, was part of it. The sports hall and its facilities for the elementary school Hudcova 35, Brno, which is the subject of the diploma thesis, is located in the development area of Brno - Medlánky. The location of the sports hall is justified due to the location of Hudcova 35 elementary school and the future development of the Medlánek area. The building is divided into a part for athletes, a part for visitors and a technological part. A parking lot and a parking lot for buses are proposed for the sports hall. The surroundings of the entire sports hall are designed for leisure activities - a workout field, a children's playground and a park. The proposal respects the environment in which the building is designed, both with the height levels of the surrounding buildings and the selected visual material, which blends in with the adjacent park and cottage development. The building meets the requirements for barrier-free accessibility and environmental requirements. In order to better blend the building with its surroundings, trees are planted in the immediate vicinity, which further form an imaginary connection to the newly created park and the current Castle Park. The facade of the building is designed with respect to the character of its surroundings. The part of the building, which is situated towards the cottage area and the park, is lined with tanned wood, and a covered walkway is designed along this part. The walkway consists of black V-shaped steel columns and a glass roof. The second part of the building is situated towards the Mechanical Testing Institute. The facade is designed from ecological mineral plaster. On this part of the building, windows are designed, which are connected in a belt using thermal insulation.
Neuropeptide Y and food intake regulation
Váchová, Barbora ; Nedvídková, Jara (advisor) ; Telenský, Petr (referee)
Neuropeptide Y (NPY) is a 36-amino acid member of NPY family. Its biosynthesis occurs in the hypothalamus and its expression also in adipose tissue was described in 2007. NPY is an ubiquitous hormone with central and peripheral effects which lead to the maintenance of homeostasis; NPY is also one of the most significant orexigenic hormones. NPY utilizes G-protein coupled receptors Y1, Y2 and Y5 for its food intake regulation; owing to serious side effects of currently used anti-obesity drugs, the ligands of these receptors (Y1R and Y5R antagonists and Y2R agonists) are considered as targeting molecules for a new therapeutic strategy for obesity treatment. This work is focused on the characteristic of NPY as an orexigenic agent and the possibility of using NPY-based drugs for obesity treatment.
Regulation and supervision of the central bank over the financial institutions
Váchová, Barbora ; Bakeš, Milan (advisor) ; Karfíková, Marie (referee)
Resumé/Summary The goal of my thesis was to clearly map the system of central bank's regulation and supervision of financial institutions through defining different entities that operate in the market and risks they take. I tried not to limit my thesis to the Czech banking market but to assess impacts of significant EC directives regarding regulation and supervision on our legal order. The reason for my research was to solve a disputable issue concerning the suitability or even necessity of the financial sector regulation, compared to the claim of some economists who incline to the concept of avoiding the financial market regulation. In my opinion, banking regulation seems to be a completely natural phenomenon with positive impact on the entire economy, which mostly arises from the bank position that has to be different from other industrial or trade companies. The thesis consists of four chapters with each chapter focusing on specific aspects of banking regulation and supervision. Chapter one defines key institutions of the financial market, which are commercial banks, other financial institutions and regulated entities in the position of regulated and supervised persons on one hand, and the regulating and supervising authority on the other hand, which is the Czech National Bank in the Czech Republic. CNB's supervision over the provision of investment services
Váchová, Barbora ; Bakeš, Milan (advisor) ; Kotáb, Petr (referee)
CNB's supervision over the provision of investment services This paper describes the Czech National Bank supervision over the provision of investment services. The starting point is the current dynamic development of the capital markets within last few years, which is characterized by interlinking markets, newly traded investment instruments and cross-border transfers of capital and risk between countries and financial sectors. The work analyzes the most important categories of investment services providers (investment firms) and finds solutions to achieve the optimal Czech National Bank supervision, where Czech law is inadequate or incomplete. There are two evaluation criteria chosen, the single capital market entities providing investment services and particular investment services provided by the investment services providers. Performing the analysis the work concludes that investment services providers intermediate the trading for customers in an adequate way and clearly contribute to the development of the financial market in the country. In the future, we can expect a higher intensity of supervision by the Czech National Bank to ensure the investment services providers' compliance with all applicable rules.
Mechanism of the inhibitory effect of ethacrynic acid on vaccinia virus growth
Váchová, Barbora ; Mělková, Zora (advisor) ; Španielová, Hana (referee)
Vaccinia virus (VACV) is globally known for its use in the eradication campaign against variola virus (VARV), the causative agent of smallpox. In 1980, WHO declared VARV as eradicated and the worldwide immunization has been stopped. After 2001, vaccination of selected groups of population was re-introduced because of concerns about variola misuse for bioterrorist purposes. Therefore, drugs effective against variola, post-vaccination complications and consequences of VACV transmission in the population, are intensively studied again. In 2009, laboratory of Dr. Mělková demonstrated that ethacrynic acid (EA) inhibited growth of VACV at the level of late gene expression. The goal of my diploma thesis was to perform a detailed study of the effects of EA on VACV late protein synthesis and on other stages of VACV replication cycle. To this end, the following methods were used: infection of tissue culture cells, plaque assay, northern blot, slot blot, western blot, direct and indirect immunofluorescence of fixed cells and fluorescence of EGFP expressed by VACV in infected live cells. First, we wanted to determine whether EA affects VACV late gene transcription or translation. Our results indicate that EA does not decrease levels of VACV late gene mRNA's and therefore, it probably affects late gene...
