Evaluation of Metabolism and Excretion of Endogenous Substances and Xenobiotics

Despite the significant progress in analytical techniques, the development and validation of methods for determining endogenous compounds is still a challenging task due to the natural presence of the analyte in the investigated biological matrix. In contrast to xenobiotics, which do not normally occur in biological material, endogenous substances represent a fundamental problem leading to the impossibility of obtaining an analyte-free sample, the so-called blank, which is used in the development and validation of a new method for the preparation of reference samples. Several generally accepted procedures to solve this problem are described, such as the use of surrogate matrices, analyte-purified matrices, or surrogate analytes; however, the used procedures do not always meet the requirements for developing a reliable analytical method according to the given guidelines and regulations. Martin Uher DAROVÁNÍ ORGÁNŮ - POVINNOST, DAR NEBO ŠANCE? Bakalářská práce Resumé Práce se snaží podat stručný přehled problematiky odběru a transplantace orgánů z medicínského i etického hlediska. V první části, po úvodu a krátkém historickém přehledu, se autor věnuje vymezení pojmů z oblasti transplantologie a toho, jaké druhy transplantace známe. Potom následuje kapitola o mozkové smrti jako smrti jedince. Popisuje postupy, jak k diagnóze mozkové smrti dospět. Důležitost důkazu smrti jedince je založena na zásadě, že není možný odběr orgánu z mrtvého. V následujících dvou kapitolách se práce věnuje jak dárcovi tak i potencionálnímu příjemci orgánů - jaké "podmínky" musí splňovat a jaké orgány může darovat, resp. přijmout. V 6. kapitole autor popisuje vlastní výkon transplantace a 7. kapitolu věnuje velmi aktuálnímu problému - transplantaci od žijícího dárce. Další, 8. kapitola se specifickými problémy týkajícími se transplantací - jako jsou např. dárci s nebijícím srdcem, děti a anencefaličtí novorozenci - uzavírá "medicínskou" část práce. Autor se dále věnuje etickým aspektům transplantace - od diskusních rovin přes etické principy až po problém obchodu s orgány a etické problémy darování orgánů od žijících dárců. MicroRNA expression in glucocorticoid-treated patients with systemic autoimmune

Rheumatoid arthritis is the most common joint disease of autoimmune origin. It is accompanied by inflammatory conditions that lead to irreversible changes in the joints, their deformities ending with permanent disability. Treatment of the disease involves routine regimens, surgical, as well as pharmacological treatment, which is necessary for advanced forms. Glucocorticoids play an important role in the therapeutic intervention in the course and progression of the disease. In spite of their anti-inflammatory effect, which is a key to improving the condition of the patient, they have a number of side effects in the long term- use. In this study, we have focused on the impact of these drugs on microRNA expression changes in arthritic patients treated with pulsed doses of glucocorticoids. MicroRNAs are nowadays widely studied due to their possible use as biomarkers in monitoring disease progression and the effect of treatment. MiRNA expression analysis was performed by quantitative real-time PCR array of 754 miRNAs with reverse transcription using stem-loop primers that allow amplification of short sequences that microRNAs are. Swing option valuation on electricity and natural gas markets

Swing options had been part of natural gas market before its embedded option feature was appreciated. The flexibility of delivery is valuable because of characteristic features of energy commodities as non-storability, high frequency of events and seasonality. Swing options enable this flexibility. Holder of the option is allowed to react to market situation in flexible way and change the amount of delivery up or down in some known intervals. Total deviation from negotiated amount can't exceed some boundaries in case of "take-or-pay" condition. It is not unique general valuation form of such flexible contracts as swing options. General definition of Longstaff Schwartz Least Square approximation method (LSM) is provided at first. Then it is shown other standard valuation concept as finite difference method. It is also mentioned tree method and more complex dynamic stochastic programming method. Analysis of energy commodities time series of central Europe is done and it is shown example of LSM approach use in valuing swing option with underlying asset of base load electricity in Czech Republic.
