DNA isolation from probiotic lactic acid bacteria in food additives

In this work the functionalised magnetic particles were tested with streptavidin to selective DNA isolation. The method of selective DNA isolation was tested by using DNA probiotic strain Lactobacillus paracasei subsp. paracasei CCDM 211/06. A test was done on the biotinyl oligonucleotic particles, which was immobilised by containing streptavidin and it was used like a DNA probe for isolation complementary DNA chain by means of DNA/DNA hybridization. The primer R 5´ bio and the biotinyl denatured specific PCR product were tested for species Lb. paracasei as a DNA probe. These following experimental conditions were optimized for selective DNA isolation: temperature and time of hybridization, amount of DNA and the release of DNA from microspheres. Isolation of DNA was verified by PCR with specific generic primers. The specific generic PCR product was amplified in extent 250 bp, which was detected by using electrophoresis in agarose gel. This optimized method was successfully used in selective isolation of DNA Lactobacillus from a complementary sample of supplementary food (BIFI pangamin).
Voltammetric Determination of Dinitronaphthalenes Using Single Crystal Silver Amalgam Electrode

Voltammetric methods for determination of 1,3-dinitronaphthalene (1,3-DNN), 1,5-dinitronaphthalene (1,5-DNN) and 1,8-dinitronaphthalene (1,8-DNN) using DC voltammetry (DCV) and differential pulse voltammetry (DPV) at novel single crystal silver amalgam electrode (CAgAE) were developed in this work. Developed methods may be used for determination of 1,3-DNN within a concentration range 2 - 10 μmol.L-1 with LoQ 2 μmol.L-1 using DCV and 1 - 100 μmol.L-1 with LoQ 1 μmol.L-1 by DPV; 1,5-DNN wihin a concentration range 2 - 10 μmol.L-1 with LoQ 2 μmol.L-1 using DCV and 1 - 10 μmol.L-1 with LoQ 1 μmol.L-1 by DPV and 1,8-DNN within a concentration range 2 - 10 μmol.L-1 with LoQ 2 μmol.L-1 using DCV and 0,3 - 1 μmol.L-1 with LoQ 0,3 μmol.L-1 by DPV. An attepmt to decrease LoQ by adsorptive stripping differential pulse voltammetry was not successful. Process of electrochemical reduction of selected dinitronaphthalenes on the working electrode was also studied by cyclic voltammetry. This work verified the use of CAgAE as alternative electrode material to mercury electrodes for determination of electrochemically reducible organic compounds in aqueous-methanolic media. Legal regulation of the liquidation of a business company

Resumé Liquidation of a company is the process whereby the life of a company is brought to an end and by which its property should be administered for the benefit of its members and creditors. Most of the liquidations occur because a company has cash flow problems and can't pay its debts. The liquidation of a company occurs in the Czech Republic between the winding-up and dissolution of a company. The legal regulations of liquidation can be found in the Commercial Code (Act No. 513/1991 Coll., as amended). A company becomes dissolved on the day it is deleted from the Commercial Register. The dissolution of a company is preceded by it's winding-up. A company should be wound-up with the liquidation, unless the law provides for otherwise. A company comes into liquidation at the day it is wound up. During the period when the company is in liguidation, it's commercial name shall be followed by the words "in liquidation". The main person who takes control of the company is the liquidator. The powers of the statutory organ to act in the name and on behalf of the company shall pass to the liquidator on his appointment. The liquidator shall execute only legal transactions which relate to the company's liquidation. Liquidation shall be completed by distribution of the liquidation remainder. After completing...
