The Influence of the Size and Shape of Test Specimen on the Elastic Modulus of Lightweight Concrete

The diploma thesis deals with the determination of the influence of size, shape and type of test specimen on values of modulus of elasticity of light-weight concrete. A lot of different specimens were prepared from two concrete’s mixtures for the experiment. Tests for measurement of static modulus of elasticity and dynamic modulus of elasticity by ultrasonic impulse velocity method were made. The results were assessed and summarized in tabular and graphical form.
The Modulus of Elasticity of Air-Entrained and Non-Air-Entrained Concrete

The bachelor thesis deals with the verification of the degree of influence of the air-entraining admixture on the modulus of elasticity of concrete. The experiment was carried out for these purposes. The values of static and dynamic modulus of elasticity were determined on drilled cores from concrete blocks of different mixtures. The magnitudes of dynamic modulus of elasticity were determined by using an ultrasonic pulse velocity method and a resonance method. The results are summarized in tabular and graphical form.
Heme sensor proteins as potential biomarkers of cellular and oxidative stress processes induced by ionizing radiation

[IN CZECH] Ionizing radiation is a potential inducer of the oxidative stress processes in cells. As a result, reactive radicals are formed in the intracellular space modifying the essential biomolecules. Ionizing radiation has either direct (radiation sickness) or indirect (malignant processes) effect on the organism. Therefore, a fast determination of the dose is required when suspected irradiation of the organism occurs. However, a method routinely applicable, fast enough and at the same time suitable for dose estimation based on biomarkers has not been developed so far. The aim of this thesis is to describe new properties of the selected heme sensor proteins and discuss their potential importance for the cellular adaptation to oxidative stress conditions. Specifically, the thesis is focused on two eukaryotic proteins, heme regulated inhibitor (HRI) and transcription factor p53. The study of functional regulation as well as the conformational changes of these proteins induced by heme is greatly emphasized. Besides, the optimization of the key experimental methods was conducted. In this study, a panel of eight different PCR-ribotypes (RTs) with their proteins released in vitro were subjected to analysis. The aim of this work is to monitor the relationship between secretions of individual proteins associated with flagellar formation and function in C. difficile strains of variable virulence. Methods: Within our research, a combination of tandem mass spectrometry with liquid chromatography was used. The semi-quantitative analysis employed label free quantification (LFQ) approach. Results: From the quantifiable proteins, 17 were significantly increased in functional annotations. Jana Netriová, PhD. Title of diploma thesis: Laboratory diagnostics of kidney injury in patients with diabetes mellitus I and II Diabetes mellitus is a chronic metabolic disease. This disease leads to develompement of both microvascular and macrovascular complications. In this work we examined a microvascular complication - diabetic nephropathy - and its laboratory diagnostics. We focussed on parameters cystatin C and creatinine. We estimated a glomerular filtration with Grubb, Stevens and MDRD equations. The Grubb and Stevens equations use cystatin C to determine the glomerular fitrlation rate (GFR), MDRD equations use creatinine, albumin and urea. For the asessment of kidney function we evaluated albumin and total protein waste in urine samples collected for 24 hours. The parameters were measured in file of 183 patients. Number of pacients with diabetes I.type was 49 and with diabetes II.type was 134. In Vitro Study of Anticancer Effect of Amaryllidaceae Montanine Alkaloids

The possibilities of current therapy of cancer diseases are wide, yet these diseases are still among the most common causes of death. The current findings of biomedical research are increasingly being applied in the treatment strategy of cancer. Understanding the molecular pathways associated with cancer cell formation, progression, and metastasis has led to the discovery of new anticancer drugs and the development of targeted therapies. Simultaneously, knowledge of the molecular mechanism of action of anticancer drugs derived from plants or their semisynthetic derivatives makes it possible to expand the portfolio of cytostatics used so far. The aim of the dissertation was to study the cytotoxic effect of hitherto unexplored montanine-type isoquinoline alkaloids in vitro using a panel of cancer cell lines and a non- cancer cell line of human lung fibroblasts. In the initial phase of the study, the basic types of montanine alkaloids, montanine and pancracine, were isolated, further subjected to cytotoxicity screening using a panel of 9 cancer cell lines of different histotype and one non-cancerous cell line formed by lung fibroblasts. Radiosensitization of cancer cells by modulation of autophagy: Phosphoproteomic analysis

(ENG) Lung cancer, and in particular, non-small cell lung cancer (NSCLC), belongs to the most lethal oncological diseases worldwide. Choosing the proper therapeutic approach is, therefore, inevitable for the successful treatment of lung tumors. In this regard, radiotherapy is an essential therapeutic approach. However, like any other treatment option, radiotherapy has its limitations in the form of radioresistance. Autophagy has been generally accepted to be one of the most crucial factors in the radioresistance of tumor cells. It is, therefore, necessary to search for suitable approaches that would lead to the suppression of cytoprotective autophagy and, thus, to the radiosensitization of tumor cells. In this respect, in our study, we employed methods of quantitative phosphoproteomics along with several techniques of molecular biology to examine the underlying molecular mechanisms and signaling pathways involved in the regulation and inhibition of the autophagic process.
