The Building on Obilní trh
Hladíková, Tereza ; Skála, Ondřej (referee) ; Kratochvíl, Jan (advisor)
The grain market is part of the city center of Brno. The grain market is a square with a park that overlooks the Špilberk hill. However, Údolní Street, as a connecting link between the center and the Grain Market, has several shortcomings. One of the key ones is the gap between the Ombudsman's office and the public transport stop. There is currently an unsuitable car park and several dilapidated houses on this gap. These houses are slowly being demolished due to their poor technical condition. An undeveloped gap in the Grain Market is a long-term problem that needs to be addressed. It is currently the property of the South Moravian Region. According to information, he does not currently intend to address this issue in the near future. In the future, he is interested in stopping it. This plan is in sight, but when it occurs, the City of Brno, which is interested in the plot, will also take part in this project. The building for the office of the South Moravian Region, or rather its branch, will be installed in the gap. The current headquarters does not have sufficient capacity and it completely lacks any conference center or place where unions could expand and add to more sections. The proposal and the overall idea to choose a branch of the regional office were created in consultation with the regional office. In the future, it is not expected that the current unions will disappear, but rather that sub-committees may begin to be selected and formed, thus creating the right to additional office space. The proposal also takes into account the space for catering for employees and the public. The idea of transparency allows the interconnection of individual structures. The bachelor's thesis will test knowledge in the areas of design of urban public spaces, office buildings, buildings for temporary housing, interior design and technical design of the building.

Somatic embryogenesis of conifers: metabolism and roles of RFOs
Skala, Ondřej
Raffinose series oligosaccharides (RFOs= α-1,6-galaktosyln-sach; 1 ≤ n < c. 7) represent a prominent part of soluble saccharide content in mature conifer seeds. RFOs are considered to act as the multifunctional components in plants, but their precise functions in specific physiological processes remain contentious. The present study focuses on the metabolism and role of RFOs in somatic embryogenesis in two conifer species- Picea abies and hybrid fir (Abies alba x numidica). Three basic approaches were set for achieving spruce somatic embryos (SE) with diverse levels of RFOs: a) culture cultivation with exogenous RFOs, b) culture cultivation with precursors of RFOs synthesis and the last 3) culture exposition to variegated postmaturation treatments inducing RFOs synthesis (desiccation/cold stress). Parallel combinations of stresses led to the highest RFOs levels in SE. These findings associate the RFOs levels in spruce SE with protective function within desiccation state. The final levels of RFOs in the spruce SE were also regulated by endogenous levels of myo-inositol. RFOs were rapidly degraded by SE within early phases of germination independently of the initial level of RFOs. No dramatic decrease in embryo germination was found in embryos with decreased RFOs degradation achieved by treatment...
Theravada Buddhist monks in Sri Lanka: past and present
Skála, Ondřej ; Štěpánová, Irena (referee) ; Kašpar, Oldřich (advisor)
The diploma thesis presented here informs the academic public on a specific social group of Theravadian monks in Sri Lanka. The paper focuses mainly on providing a detailed survey of the present situation of Theravadian forest monks and their relations to the main-stream modern society of Sri Lanka. The author of the thesis presents the findings, which he collected during his field research between October 2004 and September 2006. The method he had chosen, the "living-in research" enabled him to become a full member of the monastic community. Therefore, besides from being an involved observer of the examined phenomena, he was able to participate in the action himself. The synchronous aspect of this work consists in a detailed examination of the present situation of the individual subjects and relations among them. The position of a monk (bhikkhu) and that of the monastic community (Sangha) in Sri Lanka traces back to 2000 years ago and the mutual monk-laymen relations have undergone many changes. For this reason, a part of the thesis is dedicated to monitoring the development of the Sangha and its position from the very beginning to the present times. Within this part, the author goes into more depth when describing the dynamic phenomena related to the culmination of colonialism in the second half of 19th...
