HPLC determination of betamethasone and chloramphenicol using alternative stationary phases

A new procedure has been developed for separation and subsequent determination of betamethasone and chloramphenicol in pharmaceutical eye drop BETABIOPTAL ® preparation. The analysis carried out using a mobile phase of acetonitrile / water (25:70, v / v), adjusted to a flow rate of 0.75 ml / min at a temperature of 30 ř C, sample injection 5 Ll in the isocratic mode and the detection wavelength of 241 nm. Internal standard with similar properties as the two active substances has been selected as propylparaben. There was chosen for the separation column Synergi Fusion RP MS 2 cm × 2 mm, with a particle size of 2 m. The peaks have sufficient resolution and symmetrical shape and one analysis lasted up to 2 min while maintaining the quality of measurements (accuracy and reliability). The optimized method was validated before the actual measurement.
Nursing care of a patient after a bariatric surgery

Obesity belongs to the category of lifestyle diseases. It is expected that the obesity incidence will continue to grow. Currently there are several alternatives of treating this disease. Every patient must go through a conservative way of treatment at first and only in case of its failure the bariatric and metabolic surgeries take their turn. This bachelor thesis was drawn up in a form of a case study which concerns a female patient suffering from almost 2. class obesity. It describes 32 year old female patient who decided to reduce her weight and underwent a gastric sleeve surgery. Taking into account the characteristics of the medical intervention the multidisciplinary team focuses its activities mainly on pre- and post-operational matters which closely relate to formation of so called non-compliance. When, however, everything goes well and there are no complications thereafter the quality of life of a patient grows in most of the cases. Separation and Determination of Betamethasone and chloramphenicol in multicomponent pharmaceutical preparations using a monolithic columns coupled to sequential injection system

SEPARATION AND DETERMINATION OF BETAMETHASONE AND CHLORAMPHENICOL IN MULTICOMPONENT PHARMACEUTICAL PREPARATIONS USING A MONOLITHIC COLUMNS COUPLED TO SEQUENTIAL INJECTION SYSTEM Markéta Salabová Abstract A new separation method based on a reversed-phase sequential injection chromatography (SIC) technique was used for simultaneous determination of chloramphenicol and betamethasone in pharmaceutical eye drops in this contribution. Short monolithic column coupled with a sequential injection analysis (SIA) system enabled separation of two compounds in one step. A Chromolith Flash RP-18e, 25-4.6 mm column with 5 mm precolumn (Merck, Germany) and a FIAlab 3000 system (USA) with a 6-port selection valve and 5 ml syringe were used for sequential injection chromatographic separations in study. The mobile phase used was acetonitrile-water (30:80, v/v), flow rate 0.48 ml min-1 ; UV detection was at two wavelengths 241 nm and 278 nm, (absorption maximum of betamethasone and chloramphenicol). The basic validation parameters showed good results: linearity of determination for both compounds including internal standard (propylparaben) > 0.999; repeatability of determination (RSD) in the range 0.8 - 1.7% at two different concentration levels, and detection limits in the range 0.5 - 1.0 µg ml-1 .
