Monika Kuchařová, Ph.D. Title of Diploma Thesis: The fluctuation of fatty acids levels in the tissues of suddenly deceased persons and comparison with values in pathological states Fatty acids and their metabolites are significantly involved in many physiological and pathological processes. This thesis monitors the levels of selected fatty acids in human tissues and the effect of long-term disease on their stores. In addition to the general characteristics, the theoretical part focuses on the intake of polyunsaturated fatty acids and their importance in the human body. It also deals with the effects of eicosanoids and other fatty acid metabolites. Methods used in fatty acid bioanalysis are also described. In the experimental part, the proportion of fatty acids in seven tissue types was compared between two groups of donors. One of them included 8 relatively young, healthy, suddenly deceased individuals. The second group included 12 elderly polymorbid patients. Monika Kuchařová, Ph.D. Title of Diploma Thesis: The determination of fatty acid levels in the tissues of healthy, suddenly deceased persons and polymorbid patients The aim of the thesis was to determine the levels of 14 fatty acids in tissues of suddenly deceased, otherwise healthy individuals and in the group of polymorbid, chronic patients. It was a comparison of fatty acid levels in seven tissues of the human body: subendocardial left ventricular tissue, liver parenchyma tissue, kidney cortex, adrenal tissue, skeletal muscle, abdominal subcutaneous adipose tissue, and brain tissue. Each group included 10 deceased patients. The theoretical part incudes fatty acids as the main component of lipids. It deals mainly with the group of polyunsaturated fatty acids and their relation to pathologies in the human body. Title of Diploma Thesis: Determination of the fatty acids spectrum in patients undergoing treatment with taxanes The diploma thesis deals with the determination of fatty acids spectrum in erythrocytes in patients treated with taxanes. The theoretical part describes general features of fatty acids, their synthesis and degradation. Then it is focused on the topic of breast cancer, taxane treatment and negative side effects associated with taxane treatment, especially polyneuropathy. The gas chromatography, which is commonly used in practice, is also described in this part. The experimental part is divided into three parts. The first part describes working process, chromatographic analysis and evaluation of this analysis. In the second part there is comparing of spectrum of fatty acids of patients without and with polyneroupathy. PharmDr. Veronika Nováková, Ph.D. Student: Kateřina Pekařová Title of Thesis: Cationic surface active compounds and evaluation of its properties Quaternary ammonium salts are used as surfactants in many branches of the food, pharmaceutical and chemical industries. The basic properties of these substances were studied in the thesis. The critical micellar concentration of the homologous series of five substances was determined using the conductometric method. The theory of declining critical micellar concentration with the increasing length of the side alkyl chain has been verified. The hydrolytic activity of two commonly used cationic surfactants benzalconium chloride and benzoxonium chloride for the decomposition of the organophosphorus pesticide fenitrothion was tested. For these two substances, the effect of pH and temperature changes was evaluated. The influence of tenside concentration was also monitored. Benzoxonium chloride at 50 řC and pH 11was found as the most effective hydrolytic catalyst for the decomposition of the organophosphorus pesticide fenitrothione.

Martin Drastík, Ph.D. Title: Stevia rebaudiana - food and pharmaceutical industry applications The first part of this thesis deals with the description of the plant and its substances, the most important are steviolglycosides, especially stevioside and rebaudioside A. In the following chapter, the toxicity and safety are evaluated, which is directly related to the use of stevia and steviolglycosides in food and pharmaceutical practice. In the next part, the utilization in the food industry and connected legislation is described. The harmfulness of the refined sugar is outlined as well. Application in the pharmacy is not so widespread, therefore this chapter deals primarily with the effects of steviolglycosides. Stevia rebaudiana Bertoni and steviolglycosides have great potential to replace sugar and other sweeteners. Due to their beneficial effects, they are very popular among researchers, so we can look forward to their widespread application in food and pharmaceutical industry in the near future. The effect of cosmetic products with different pH on the skin properties of men and women

The aim of this diploma thesis was to summarize the basic knowledge of human skin and it's biophysical parameters. Attention was focused on influence of cosmetic products with different pH on the change of skin parameters especially. The first part describes the anatomical and histological structure of the skin, it's basic functions and development. In the following chapters of this work is mentioned physiological representation of microorganisms found on the skin surface and general characteristics of cosmetic products. There are used in everyday skin care that affect the natural skin surface pH value. Optimum surface pH is important for the proper functioning of human skin. The final chapters of the described the characteristics of the basic biophysical parameters of the skin (hydration, TEWL, pH, sebum) and methods for their measurement. The influence of spinal anesthesia on the degree of DNA damage

Background: The human organism is exposed daily to many endogenous and exogenous substances that are the source of oxidative damage. Cell structures, including DNA (deoxyribonucleic acid) in the nucleus are damaged due to high concentrations of these substances and accumulation of oxidative stress in cells. The predominance of these damaging processes may later be responsible for human diseases such as cancer, neurodegenerative diseases or heart failure. In our study, we observed oxidative damage at the DNA level due to spinal anesthesia. Methods: Sample processing was performed by comet analysis. The principle consists in fixation of cells (lymphocytes) in the agarose gel, lysis of cell structures for nucleotide release, incubation with specific enzymes and exposure to electrophoresis. Damaged, negatively charged parts of the DNA in the electric field are directed to the positive charged anode, creating a typical comet shape. For visualization, the gels were stained with ethidium bromide (DNA intercalating dye). Results: We have quantified single-strand breaks, oxidized purines and pyrimidines (use of enzymes to detect specific damages). The results are reported in percentage of DNA in the comet's tail.
