The aim of this thesis is to statistically compare the two methods and perform an evaluation of the data obtained, to assess whether the two methods provide similar results and are interchangeable, or whether a statistically significant difference can be observed. This diploma thesis mainly focuses on inhalant allergens of moulds, mites, house dust mites and animal allergens. The theoretical part of this diploma thesis is focused on allergies and their clinical forms. Karolína Jankovičová, Ph.D. Title of diploma thesis: Monitoring of sublingual allergen immunotherapy using serum levels of allergen specific IgG antibodies The first aim of the thesis was to evaluate the clinical course of sublingual allergen immunotherapy (SLIT). Another aim was to assess the changes in antibody response over time and to compare the results of the Oralair treated group with the control group. Last but not least, the aim of the thesis was to evaluate the changes in the levels of specific IgG antibodies in relation to the clinical course of SLIT. The visual analogue scale (VAS) was used for the clinical assessment of allergic symptoms. A commercially available ELISA method was used to determine sIgG4 against grass pollen allergens. sIgG1, sIgG2 and sIgG3 were determined by an in-house developed and validated ELISA. Based on the results, statistical analysis was performed. It was found that Oralair therapy was clinically effective from the first season of the treatment. SARS-CoV-2 in Care Home Břevnice

The aim of this diploma thesis was to describe information about SARS-CoV-2 virus and coronavirus disease (COVID-19), to evaluate the course of the disease in the Home with a special regime in Břevnice, in which the epidemic took place at the beginning of the first wave of COVID-19 pandemic in spring 2020. Another goal was to perform an antibody analysis and evaluate the obtained data. I divided the diploma thesis into three main parts: theoretical, experimental and discussion. In the theoretical part, I worked with the literature and described information related not only to COVID-19, but also to other serious infections caused by human coronaviruses. In the experimental part, which I performed in the immunological laboratory of the Department of Joint Laboratories at Havlíčkův Brod Hospital, I dealt with the issue of the clinical course of the disease, the severity of the disease and possible consequences after the infection. I also dealt with methods for the determination of antibodies, the principles of which are described in Chapter 4.5. The diagnostic methods and procedures used are described in Chapter 5.3 Laboratory Assays. Vladimíra Vroblová, Ph.D. Title of diploma thesis: Comparison of the immunophenotypes of normal and pathological plasma cells - determination of the rate of risk of progression in patients with MGUS Four-color flow cytometry is a necessary part of plasma cell imunophenotypisation today. It allows to define plasma cell count with a high accuracy and to distinguish normal and pathologic cell on the base of surface and cytoplasmatic markers expression. An expression of the following surface markers, CD138, CD38, CD19, CD56, CD45 CD20 and CD27, was monitored in this dissertation. An expression of the cytoplasmatic light chain κ and λ followed which confirmed monoclonality of the plasma cells in patients with multiple myeloma or with monoclonal gammopathy of undetermined significance. In reference group of patients the expression of light chain κ and λ confirmed polyclonality. 47 samples of patients with diagnosed multiple myeloma or with monoclonal gammopathy of undetermined significance and 22 samples from patients with increased count of plasma cell, but without present hematological disease of plasma cells,...
