Světlák and of the author P. Zitová. The diploma thesis is divided into the two main parts - the theoretical part summarized the issue of the temporo-limbic dysfunction, the research part answers the put questions. The main goal of the diploma thesis was to describe the issue of the temporo-limbic dysfunction in the university student sample divided into smokers and non-smokers.

Title: Temporolimbic dysfunction in persons with sleep disorders Background: The aim is to demonstrate the occurrence of symptoms of the temporolimbic dysfunction in person with sleep disorders using standard psychosocial questionnaires. Methods: The occurrence of symptoms of the temporolimbic dysfunction was detected by means of CPSI and LSCL-33 questionnaires. Correlation of both questionnaires' results was evaluated by Spearman's coefficient. Results: According to CPSI 11 % patients have abnormal score and they have higher occurrence of symptoms of the temporolimbic dysfunction. According to LSCL-33 achieve 32 % of respondents abnormal score and 7 % respondents have pathologic score, which means the presence of the temporolimbic dysfunction. Conclusions: The occurrence of the temporolimbic dysfunction in persons with sleep disorders have been shown, but it haven't been higher in comparison with healthy population. Sodium (Na+ ) is the major extracellular cation. It plays an important role in maintaining membrane potential and depolarization that is the basic mechanism of transmission of the nerve impulse. It is involved in maintaining acid-base balance, osmotic pressure, water retention in the body. The largest part of the sodium is in the extracellular fluid where it is stored about 50 % of sodium. Plasma concentration of sodium is 140 ± 5 mmol/l. Drug-induced electrolyte disorders are increasingly reported and may be associated with considerable morbidity and mortality. The risk of drug-induced hyponatremia is generally higher than the risk of drug-induced hypernatremia. Hyponatremia is a common electrolyte disorder defined as a decrease plasma sodium concentration below 135 mmol/l. It is classified by the state of volume - hypovolemic, euvolemic and hypervolemic hyponatremia. It reflects the relative rate between sodium and water in the body. Adéla Dolejšová Testing the efficacy of potential therapeutics for Alzheimer,s disease Charles University in Prague, Faculty of Pharmacy in Hradec Kralove Pharmacy The aim of this thesis was to find out whether already implemented Ellman's method, which is used to analyse irreversible inhibitors AChE, is going to be a suitable technique for measuring reversible inhibitors. Furthermore, the efficiency of newly synthesized AChE inhibitors was established. These inhibitors will be used for treating AD or as prophylaxis against neural paralytic substances. Moreover, it was compared affect the efficacy of AChE inhibitors after intramuscular and intraperitoneal administration. The measurements were done in vivo on potkan species Wistar. In the first experiment standard AChE (tacrin, 7-MEOTA, donepezil, rivastigmin) inhibitors were applied to verify the method. In the second experiment the newly synthesizes AChE (K 298, K 344, K 474) inhibitors were medicine. The results confirmed the effectiveness of the tested method on commonly used inhibitors such as donepezil and rivastigmin. Out of the newly synthesized K 298, K 344 and K 474 inhibitors none was proven to have any significant inhibiting activity. Consultant: PharmDr. Vendula Šepsová The aim of the thesis was to determine the type of an inhibition of newly synthesized AChEI and to find out if AChEI structure changes influence the type of an inhibition. Altogether 12 substances (7 tacrine hybrides and 5 7-methoxy- donepezil hybrids) were investigated. The inhibition potential of the tested substances was studied in vitro on the human recombinant AChE. Spectrophotometric Ellman method was utilized as the measurement tool. The noncompetitive type of an inhibition for substances EN 1-5, PC-25 and PC-33, mixed type of an inhibition for substances PC-48 and PC-49, uncompetitive type of an inhibition for substances EN-6, EN-7 and competitive type of an inhibition for the substance PC-37 was determined. The aim of this research is the occurrence of symptoms of the limbic dysfunction in patients suffering from headaches. By means of the questionnaire survey method was tested group of people with this diagnosis. The basic premise is the assumption that certain symptoms, specifically abnormalities in brain activity, occur more frequently in patients with this diagnosis. The theoretical part of this thesis concentrates on the function of the temporal lobe and the limbic system, a brief outline of the characteristics of temporo-limbic dysfunction and a summary overview of headaches. The research part is addressed to the evaluation questionnaire study and answers to the questions asked. MUDr. Radovan Malý Ph.D. Title of Thesis: Deep vein thrombosis of lower limbs - the therapy of thrombosis in women using alteplase Background: The aim of study was observed undesirable effects of treatment of deep vein thrombosis, than safety and how long lasting an intervetion, incidence of deep vein thrombosis on lower limbs, different ages and risk factors conecting with this disease too. Methods: In this study we administred Alteplase - a thrombolytic drug - directly to the vein using a catheter. Alteplase is a tissue plasminogen activator and it activates the transformation of plasminogen to plasmin and than plasmin breaks down blood clots and helps to renew the blood floow in the affected vessel. The thrombolytic drug was administered directly to the thrombus with using catether. In this study the 68 women were treated regardless of their age. Blood sample from pacients took away before, during and after inervention to set down levels of platelets and fibrinogen. Discovery of c-Jun N-terminal kinases (JNKs), involved in regulation of gene expression, inflammation, cell proliferation and apoptosis, may contribute to better understanding of AD pathogenesis. JNK3 is a brain-specific JNK isoform involved in apoptosis of mammalian neurons. Precise localization of JNK3 in the brain still remains unclear. JNK3 silencing achieved by gene knockout had directly decreased beta amyloid (Aβ) levels in previous AD models in mice. We performed an immunohistological study to localize JNK3 gene in the mice brain in order to better focus future gene targeting therapy.
