Mechanistic insights into alcohol-induced interstrand crosslink repair by the nuclease SLX4-XPF-ERCC1

Alcohol ranks among the most widely used recreational drugs in the world, even though it is considered a risk factor for more than 200 diseases. The primary negative impact of alcohol lies in its metabolite, acetaldehyde, which, as a highly reactive compound, can form mutagenic adducts and interstrand crosslinks (ICLs) in DNA. The formation of ICLs, which have a covalent nature and block the separation of the two DNA strands during replication, is one of the important causes of mutagenesis and carcinogenesis. To maintain genomic stability, repair mechanisms have evolved. One of them is a pathway that uses proteins encoded by Fanconi anaemia genes, whose defects lead to the disease of the same name. Defects in repair pathways can be particularly dangerous in individuals with impaired functionality in other metabolic pathways, such as alcoholics and individuals with mutations in genes that result in the accumulation of toxic acetaldehyde. The theoretical part of this thesis deals with alcohol metabolism, in vivo acetaldehyde formation, and its interactions with DNA. The ICL and their repair pathways are characterized in more detail. Using computers pre-school age child

The Bachelor thesis is about aspects of computer use by preschool age child. It is explained the major role of the family and the nursery in relation to the child and the use of computer by child of preschool age. In the research part are collected opinions and attitudes of parents and nursery teachers on this issue. Selected results on introduction children with the basics of safe use of computers at home and at the nursery are confronted with the recommendation of best practices. The objective of this thesis is to introduce the use of computer child of preschool age, from the parents' perspective and teachers of the nursery. HAVLÍKOVÁ. J. Modern means of payment. Bachelor thesis. Brno: Mendel University, 2017. This bachelor thesis is focused on the analysis of the use of modern means of payment in the Czech Republic. The literary overview defines basic terms such as payment system, CERTIS and subsequently individual means of payment. The empirical part consists of two analyses. The first one is focused on supply which is represented by banks providing services to retail clientele without branches of foreign banks and specialized banks. The second analysis is intended for demand and this side consists of persons over the age of 18. There are suggested recommendations on improvement of the modern means of payment based on the accomplished analyses.

TITLE: Orientation in time at kindergarten focusing on the Sun and Moon AUTHOR: Bc. Jana Havlíková DEPARTMENT: SUPERVISOR: PhDr. Micheala Kaslová ABSTRACT: This diploma thesis deals with the development of orientation in time focusing on the Sun and Moon in preschool children as part of the attendance at kindergarten. In the theoretical part, in addition to the definition of the term of time from various disciplines, there is also an outline of the development of perception and measurement of time in terms of both evolutionary and individual development. Many activities in kindergartens are preserved for a long time, however the observation of the sky itself has slowly disappeared from education programs and plans. The practical part is focused on setting up scenarios of individual activities for observing the position of the sun in the sky, the length and direction of the sun's shadow, and the awareness of the alternation of the day with the night and the seasons. All this within the framework of action research, which was implemented in one kindergarten class. Growth of human adipose-derived stem cells on Ti-6Al-4V alloy with various surface modifications

Adipose-derived stem cells (ASCs) are of a great importance for bone tissue engineering. We compared ASCs obtained by liposuction under two pressures (-200 mmHg and -700 mmHg). The number and proliferation activity of cells isolated under -700 mmHg (ASC-H) were higher than in cells isolated under -200 mmHg (ASC-L). However, the ASC-L was more active in osteogenic differentiation, as manifested by a higher intensity of fluorescence of alkaline phosphatase and osteocalcin in these cells. The adhesion and growth of ASCs were then studied on Ti-6Al-4V samples either unmodified (K, roughness parameter R-a = 280 nm) or modified by shot blasting and tarnishing A, R-a = 200 nm), vibratory finishing (B, R-a = 100 nm) and vibratory finishing, shot blasting and polishing (C, R-a = 80 nm). All modified samples were more wettable than the K samples. On day 1 after seeding, the size of cell spreading area on some modified samples was lower than on K samples, namely on B samples in ASC-L, on B and C in ASC-H and on A in control MG-63 cells. On day 3, the cell number on some modified samples became higher than on K samples, namely on C samples (ASC-L), on B (ASC-H) and on B and C s (MG-63 cells). On day 7, the numbers of ASC-H and MG-63 cells on all modified samples evened out and became significantly higher than on K samples. On ASC-L samples, the highest cell numbers were obtained on A samples. Thus, all studied modifications of Ti-6Al-4V enhanced growth of ASCs and human osteoblast-like MG-63 cells.
Non-Romani inhabitants of socially excluded ?Romani? localities

This diploma thesis with the title ?Non-Romani inhabitants of socially excluded ?Romani? localities is divided into two parts: the theoretical and the empiric parts. The theoretical part of the diploma thesis deals mainly with defining the terms of social exclusion and socially excluded locality. At the same time, it describes the phenomenon of the poverty, underclass and white trash culture and ethnicity. The research part deals with the description of the actual research methodology, interpretation and the presentation of the research investigation results. The aim of the diploma thesis is to describe the perception of life in a socially excluded ?Romani? locality, by non-Romani inhabitants. With respect to the aim of the diploma thesis I chose the qualitative research strategy. The qualitative investigation was conducted by the method of questioning. The questioning technique was a semi-structured interview consisting of open questions. The target set consists of twelve respondents living in three excluded localities in the territory of the town of České Budějovice, who do not consider themselves, on the basis of self-identification, as members of the Romani ethnic group. The results of my research investigation shows what leads non-Romani inhabitants up to living in socially excluded localities, if they feel safely there and how they perceive coexistence with Romani ethnic group.
Harmonization of the Czech accounting legislature to the international accounting

This thesis concerns the issues of the harmonization of the Czech accounting legislature on the international accounting. The thesis is divided into two parts. The first part defines the fundamental principles of the International Accounting Standards and compares them with the accounting legislature of the Czech Republic. It outlines the basic responsibilities for the accounting. Further, it compares the conceptual framework and the valuation bases. One of the chapters deals with the entering of the International Accounting Standards into the Czech environment. The second part concerns the analysis of Czech accounting in relation to the specific International Accounting Standards, in particular, IAS 1 {--} Insurance Contracts, IAS 16 - Property, Plant and Equipment, IAS 17 {--} Leases. Finally, the different standards are compared with the Czech accounting legislature and the IFRS Financial Statement is presented.
Opinions of teachers and pupils in the second level of basic schools in Týn nad Vltavou about legalization of marihuana

The bachelor thesis is called Opinions of teachers and pupils in the second level of basic schools in Týn nad Vltavou about legalization of marihuana. In the theoretical part the thesis contains the basic information about the history of marihuana, its effects, use and related risks. My bachelor thesis also describes the areas which take advantage of medical effects of the drug, as one of the main arguments of defenders of its legalization is the fact that marihuana can have positive effects on human health. For better understanding of the issue, the theoretical part also includes an explanation of the position of hemp-based drugs in international treaties, European legal code and in the Czech legislation. In the practical part I have used a questionnaire survey in basic schools in Týn nad Vltavou to find out opinions of pupils and teachers about potential legalization of marihuana. Apart from the opinion about the legalization as such, the questionnaire also contained questions whether the respondents consider the drug dangerous for human health and for the society and which specific complications associated with its use are known to them. The conclusion of my thesis is that pupils are slightly more benevolent to the legalization of marihuana. Eighty percent of the surveyed teachers were strictly against the legalization. One on my conclusions is the fact that the surveyed pupils support legalization of marihuana to a great extent but only a small percentage of them are interested in detail in that issue. Therefore I believe that it would be appropriate to increase the general level of information about the drug, not only among the pupils but also among their teachers.

Stocks are the important components of short-term assests. It is a case of such components of assets where single tax consumption during production process is. Stocks include material, products, goods etc. There are two ways of stocks records all accounting entity can be used apart from audit duties. There are A, B ways of stocks record. Evaluating of stocks is the important component for the company. There are ways of stocks evaluating belong the method of weighed aritmetical mean variable, method FIFO and the method of fixed stock price. Individual techniques of stocks evaluating have different influence on management result. The significant criterion in the selection is mainly practicality and propriety of techniques evaluating. This criterion influences above all the size of accounting entity and stocks quantily. One of the main aims of the company are the lowest material costs result in lower selling price. All the same time the company tends to the lowest tax basis. On the contrary the highest material costs help this process.
