How to meet needs of seniors in hospital care.

The submitted dissertation of the PhD study programme Longevity Studies was prepared and financially supported by the project GAUK - Grant Agency of Charles University No. 760219 entitled "Met and Unmet Needs Of Particularly Vulnerable Older Patients in Home and Inpatient Care", of which I am a co-investigator. The mentioned research project is also in the intentions of the strategic plan and long-term activities of the research team CELLO (Center for the Study of Longevity and Long-Term Care), which is the scientific basis of the PhD study programme Longevity Studies. The main goal of the GAUK project is to provide a comprehensive overview of the needs of vulnerable older patients who are provided health care. The project is focused on three areas, which are: the needs of patients with dementia, the needs of geriatric patients at home and the needs of geriatric patients in hospital care. Partial goals are the exploration of current knowledge of the issue based on a review of Czech and foreign literature. The aim of the survey is to find out attitudes and opinions on satisfying needs from the perspective of patients and healthcare professionals. The oxidative DNA damage and its relationship to the cytotoxic effect.

This work has been focused on the study of oxidative DNA damage induced with hydrogen peroxide and on the relationship of this damage to cytotoxic effect . The repair of the oxidative DNA damage and the cell survival was followed both in normal Chinese hamster cells and their mutant derivative UV-20. The modified single cell gel electrophoresis (the comet assay,) was used to estimate the DNA damage. The cytotoxiciy was measured using the inhibition of colony forming capacity of cells. Both normal AA8 and mutant UV-20 cells repair the oxidative DNA damage with approximately the same velocity, and they show also the same sensitivity towards the hydrogen peroxide. These results indicate, that the defect in the nucleotide excision DNA repair in UV-20 cells does not play any important role in the response of these cells to the oxidative DNA damage, and therefore this mechanism is not involved in the removal of this damage. This may indicate, that the main mechanism of the repair of oxidative DNA damage is the base excision repair. Keywords: Oxidative DNA damage, hydrogen peroxide, comet assay, excision repair.
The integration of disabled people into the society

This bachelor thesis concerns itself with disabilities, disabled people and their social integration. The theoretical part deals with the various types of disabilities, how the intact public views people with disabilities and the evolution of their perception from the past to the present. It's mainly focused on education and subsequent employment of the handicapped and the approach of the government and the role it plays in the lives of the disabled. The practical part ties into the findings of the theoretical part, its goal is, by distributing a questionnaire, finding the level of knowledgeability of the intact public about people with disability, their experience and relations with them, then comparing these findings with the opinions and experiences of a few handicapped people and people with a close understanding of the issue. In the last part these findings are summarized and practical recommendations for increasing the public awareness on this topic are made.
