COHERENCE-CONTROLLED HOLOGRAPHIC MICROSCOPY IN DIFFUSE MEDIA

This thesis deals with imaging through diffuse media in coherence-controlled holographic microscope (CCHM) developed in IPE FME BUT. The mutual coherence function as well as the signal dependence on the lateral mutual shift between both arms of the CCHM are calculated. Both functions are related to each other. The latter dependence is measured experimentally. A principle of imaging with CCHM through diffuse media with both ballistic and diffuse light is explained by a simple geometrical model. This model is then verified experimentally by imaging a sample through diffuse medium. The point spread function (PSF) of CCHM for imaging through diffuse media is then calculated. Results of PSF calculation are proved experimentally.

Micropositioning tracking system

This bachelor's thesis deals with the design and construction of a micro positioning device that will monitor the movement of a robotic mouse in a defined space with the help of a camera. The main task will be to assemble a moving platform, the aim of which will be to be in constant contact with the robot below it and to copy its 2D trajectory. The device will then be applied to a live mouse for in vivo holographic endoscopy experiments.

Modifications in mechanical design of the coherence controlled holographic microscope

The aim of diploma thesis are modifications in mechanical design of the coherence controlled holographic microscope which will lead to reduction of some deficiencies. In this thesis there is a brief description of the history interference microscopy and differences between each types of interference microscopes. In this work there are stated some design deficiencies of actual condition of the microscope and also several solutions are suggested and shortly described. Then the best solution is chosen which is adjusted to a final form. It is described in detail and then realized. There is a summary included in the conclusion whether the new solution is suitable and if it fulfills the required condition.

Device for wavefront measurement of microscope objective lenses

The wavefront reconstruction of a light wave transformed by a microscope objective is the main subject of this diploma thesis together with the design and assembly of a~measuring device and the development of a computational algorithm. The purpose is to determine optical aberrations and to compare a quality of objectives with identical parameters. The term wavefront is explained and its description using Zernike polynomials is introduced in the first part of the thesis. The following part summarizes possible methods for wavefront reconstrucion. Two methods were chosen for experimental determination of a wavefront shape – shearing interferometry and solution of the transport of intensity equation. For each method a brief characteristic is provided together with possible applications, mathematical apparatus, image processing, computational procedure, setup description and proposition and results of experiments. The suitability of both methods for optical aberration determination and microscope objective comparison is discussed. Based on the obtained results, both methods were found to be suitable for comparison of microscope objectives. The suitability for optical aberration determination is possible with certain restrictions.

Automatization of method of surface structure measurement by means of reflection mode digital holographic microscope.

Reflected-light digital holographic microscope developed at IPE FME BUT uses off-axis holography principle and low spatial and temporal coherence illumination. Microscope allows reconstruction of the image amplitude and the image phase, which can be handled in real time. The only limiting factors are imaging speed of the detector and computer performance when processing holograms. Reconstruction of image phase and amplitude allows high-resolution profilometric measurements in the vertical axis direction. This thesis deals with the automatization of profilometric measurement method proposed in [2]. Proposed method uses the combination of the image phase and the image amplitude for the measurement of specimens with surface structure the vertical size of which cause the uncertainty of the image phase by a factor of 2pí. Futher the thesis deals with the construction design of the illumination system of the microscope and its realization together with experimental verification of functionality of proposed method automatization.

Mechanical design of measurement system for reactive ion etching system

The aim of this work is to design fully working measuring system for the reactive ion etching system (RIE). The Michelson interfometer, previously developed in Ústav přístrojové techniky, v.v.i., is used in this work. The theoretical part is aimed at description of interferometric methods for precise measuring of length. In next part the etching proces with RIE is described. In practical part the testing system was constructed from the parts of Thorlabs company . The functionality was controlled with this system and the results of measuring were compared with the profilometer. In next step technical drawings were created and the whole system was made.

Microsoft Asp.NET Dynamic Data in Winforms

This bachelor's thesis describes the development of an universal application core allowing developers to build flexibile informational systems in no time. The system follows on existing technology Microsoft ASP.NET Dynamic Data and implements it's ideas on the Windows forms platform. In the opening parts of this thesis are briefly described topics such as the Microsoft .NET plaform, the Dynamic Data, or the questions of a switch-over from the web platform to Windows forms. Following parts of the thesis are dealing with the actual implementation of the example informational system. The conclusion is among other things dedicated to the analysis of an usability of the product in a real world and demands of today's market.
