Hubáček, Petr ; Cinek, Ondřej (advisor) ; Šterzl, Ivan (referee) ; Mayer, Jiří (referee) ; Žák, Pavel (referee)
Infectious complications are very common after haematopoietic stem cell transplantation (HSCT). Beside bacterial and mycotic infections, viral infections are important cause of morbidity and mortality of these patients. Among viral infections, the most common are infections caused by DNA viruses. Between January 2001 and October 2008, we have tested samples from 217 paediatric and 382 adult patients after HSCT. In total, we have tested 15 757 samples (median 31 samples for children, 20 for adult). Among them, there was 14 867 samples of peripheral blood and 890 samples of other biological materials (urine, stool, CSF etc.). Quantity of HSV, VZV, CMV, HHV-6, HHV-7, EBV, adenoviruses of group A-C, BKV and albumin gene was tested using quantitative real-time PCR technology. Results of CMV, HHV-6, HHV-7 and EBV were normalized to 10 000 human genome equivalents. HSV and VZV were detected in minute number of samples. Threshold for virostatic therapy was assessed to 100 normalized viral copies for CMV. This level was crossed in 55 children and 123 adults. Despite the virostatic treatment, symptoms of CMV infections developed in 68 patients. CMV disease, alone or in the consequence of other post-transplant complications, was observed in 20 patients, fatal in 17 of them. In 4 paediatric patients, we have detected...

Propagation of inclined cracks to the interface of ceramic laminates
Novotná, Lenka ; Trunec, Martin (referee) ; Chlup, Zdeněk (advisor)
Composite materials with laminated structure provide advantages which are utilised during component design. Low density, temperature and chemical stability are the profitable properties predetermining application of ceramic laminates. The main obstacle for wide spread of ceramic materials is their inherent brittleness. Therefore, in this thesis, the crack propagation in ceramics laminates has been extensively studied. Laminated structures with various volume fractions of components (alumina and zirconia) were prepared by electrophoretic deposition. Evaluation of crack propagation through the interface and determination of basic mechanical properties was conducted on the basis of extensive literature search. Crack deflection originated in both presence of internal stresses and differences in elastic modulus during the crack interface passing was monitored. A special type of specimen geometry was employed with the aim to set arbitrary angle between crack and interface. It was experimentally found that the degree of crack deflection is dependent on entering angle and volume fraction of components. Higher crack deflection was already found in the bulk of the test piece comparing to the test piece surface. The 3D fracture surface reconstruction generated using laser confocal microscopy was used in this detailed crack propagation study. Further basic elastic and strength characteristics of laminates were determined and compared to those obtained from monolithic materials. The validity of the mix rule for elastic characteristics was confirmed by comparing of elastics modulus. The most reliable method for elastic modulus determination was marked the dynamic resonance method due to low scatter and consistency in measurement. The flexural strength of all laminates tends to be close to the flexural strength of the weakest component. Therefore the mix rule is not applicable for flexural strength estimation on the contrary of elastic characteristics. The change of component volume fraction leads only to change of flexural strength scatter. Thanks to gained knowledge about crack propagation and basic characteristic determination will be possible to design ceramic laminates more efficiently for given needs of application.

Halbrštátová, Lada ; Havířová, Zdeňka (referee) ; Lavický, Miloš (advisor)
Abstract The main purpose of creating this object was to create a place where people having bad living situation will meet people who live regular lives. This is the reason why two floors were projected. The first floor is designed for social activities whereas the second floor is a housing part with apartments. The advantage of irregular but compact shape of the building is perfect connection between its functional parts. The object does not have any basement and it is placed in a little slopy bulit-up surroundings on the crossing of Tyršova and U Distance streets in Letohrad town. On the first floor there is a main entrance, a coffee, massages, a recreation hall, an art working-room, a gym, changing rooms, administration and the technological background of the object. The second floor does not cover a whole area of the first floor. Apartments, laundry room and drying room are situated here. One of the apartments here is projected as barrier-free as well as whole first floor. Both floors are connected by central staircase with shape of „U“ and an elevator. Roofing of the object is realized by single layered flat roof with classic layer order. The gym is roofed by a flat roof held by glued laminated girders. The construction was projected with regard to synergy of all used materials and systems. The chosen solution of object with two floors (both above the terrain) makes the building pretty marked when looking at it’s location, however it makes the place more integrated and uniform.
