Cultivation of grapevine in conditions in vitro

Grapevine is a traditional crop in the Czech Republic, especially in the South Moravia region. It is a vegetatively propagated crop; hence vineyards are endangered by abiotic and biotic stress influence. The risk of plant damage due to frost and pest influence occurs in the field conditions according to season and locality. Cultivation of grapevine in conditions in vitro eliminates the risk. Higher utilization of the method is limited by sensitivity of some genotype to in vitro conditions. This methodology solves these problems and describes material introduction into in vitro conditions, explants multiplication and slow growth condition establishment. The Ministry of Agriculture of the Czech Republic is the user of this methodology and it will utilize it in the framework of “National Programme on Conservation and Utilization of Plant, Animal and Microbial Genetic Resources for Food and Agriculture”.

Legal consequences with respect to the breach of contract under civil law

- 54 - 6. Resumé Consequences of breaking the contract in civil law - Most of the relations is in civil law established by the contract. Contractual freedom is one of the basic freedoms provided by modern democtratic state. But unfortunately with the increasing amount of contracts there is also increasing number of cases when the contract is breached (intentionaly or not) by one of the contractual partners. The law presumes such cases and provides particular consequences. These consequences are the subject of this thesis. According to the Czech civil law there are following istitutes which can follow after breaching the contract: Default, the interest on the sum in arrears, a late payment charge, damages, penalty clause, liability for defects. The thesis is divided into several parts according to the described consequence. At the beginning of the thesis, there is an introduction into the problem, then there is described the contract. There is explained the term, then the process of making the contract (proposal and its acceptation). After clarifying the basic term of the thesis follows the description of mentioned consequences. Jakubcová, N. The quality of spa and medical tourism. Bachelor thesis. Brno: Mendel University, 2015. Bachelor thesis deals with quality of spa and health tourism in Slovakia. First of all theoretical knowledge related to the issue of bachelor thesis are processed. Our work is based on the primary and the secondary marketing research. Subsequently our thesis processes results of marketing research, analyses current condition of spa, certifications and organizations functioning in the field of spa in Slovakia. The main aim of bachelor thesis is to determine current satisfaction with quality of spa in Slovakia. To evaluate quality of spa and health tourism. Spas are evaluated from the tourist point of view. In conclusion thesis proposal is created to improve the quality of services in spa and medical tourism in Slovakia.

Hypoglycaemic conditions requiring rapid arrival of emergency services in South Bohemia

This bachelor thesis deals with the hypoglycaemic states that is not only a complication for diabetes mellitus disease but there are other diseases such as Adisson's disease or congenital hyperinsulinism. Hypoglycaemia can be for some people life endangered state, mainly if the state is not recognized in time or if an effective therapy is not started. The rapid therapy is crucial and should be started within the first speared symptoms to prevent the state get worse. The thesis acquaints the reader with the major gland's anatomy that influences the glycaemia, then the pancreas and its function and its hormones function. The major hormones are insulin and glucagon. The both hormones have the main influence on the controlling of glycaemia in the body. However, they are not the only ones factors that influence the blood sugar level. In various literature sources, the different glycaemia values can be found. That is the reason why it is difficult to define the level of hypoglycaemic stat. The thesis includes several definitions of the hypoglycaemia that differ according to the physiological values mentioned in various sources. An important part of the thesis is the chapter about the clinical symptoms. In the chapter, most sources concur. The clinical symptoms can be varied, sometimes not enough specified. After a few first vegetative symptoms (unrecognized hypoglycaemia syndrome can missed them), disorders of consciousness from the conscious to the coma usually appears. The coma is a state when a patient does not react on the surrounding. The following chapter describes the process of hypoglycaemic diagnoses. The process is described both in health institutions and during the pre-hospital urgent care where the possibilities are restricted. The care is a necessary part of the state. If the patient is conscious and is able to cooperate, some sweet drink and some chocolate are the best opportunity for the first help. The ideal tool that could be used during the light type of hypoglycaemia is Glukopur. Glukopur is the pure powder glucose that helps to the body that do not have to split disaccharides as within the sweet drinks and bars. Another topic is the dangers of hypoglycaemia. The dangers include the development of dementia that is often connected with hypoglycaemic states, the brain's damage or the dangerous of the heart arrhythmia. The most serious danger that can appear is the death. The thesis describes the summary of medicaments that can cause hypoglycaemia. Hypoglycaemia can appear as a child inherited metabolic disorder or as hyperinsulinism. One of the chapters deals with this topic. The quantitative method data collection was used for the practical part of the thesis. The departures of The Emergency medical service in South Bohemian Region to the hypoglycaemic states during the year 2014 were searched. From the whole set of cases eight of them were randomly chosen and were described case interpretations that show the way of the emergency service intervention within the hypoglycaemic states. The case interpretations depict the patient's state before the arrival of the emergency service, the way of treatment and the patient's state after it. The evaluated indication by health operation centre is depicted in the following section. These indications do not have to be identical to real patient's state. One of the aims of the thesis was to map the most common factors of chosen hypoglycaemic states that were measured according to the NACA scale, grades IV.-VI. The data were gotten by the crew of The Emergency medical service in South Bohemian Region. The next one aim was to map the indications of the health operation centre and the following patient´s state according to the NACA scale.

Survey and assessment of the National museum building in Prague

The report presents results of a complex survey of the health conditions and damages and failures of the building of the National museum in Prague. It further summarizes scientific investigations of the loads, namely of winds in situ as well as on models in a climatic wind tunnel where also soiling was tested. The results of material analyses and public attitude to soiling and failures on the façade are involved, too. The report has a rich photographic annex.

Child with Diabetes mellitus in Kindergarten

This bachelor thesis deals with inclusion specialty of diabetic child to day-care centre. It is about the case study of the boy with diagnosis Diabetes mellitus of 1. type on insulin pump. The theoretical part is about illness Diabetes mellitus of 1. type, its symptoms, therapy and complications. The therapy way by insulin pump is described more in detail. The theoretical part contains the chapters regarding the psychological impacts to ill person and its family, the need to respect of the regime precautions and the legislative conditions of the sick child integration to the day-care centre. The practical part consists of a qualitative survey, which represents the boy's casuistry with diabetes. The practical part also includes an interview with the boy, his mother and a teacher at the day-care centre. Further the practical part consists of activities, which can help the pedagogues to incorporate a disadvantaged child such as this boy to the day-care group. From the findings it is clear that an insulation pump does not have any underlying influence on day to day activities in day-care centre however it makes it easier for the family and the unwell child.

Cryopreservation of common carp (\kur{Cyprinus carpio} L.) sperm under different freezing conditions

In the present study, we examined several cryoextenders previously used by several authors and various freezing protocols to determine the relative importance of each parameter on sperm freezing. The effects of controlled seeding and changes in cooling rate at different stages of freezing were also examined. Sperm samples from seven individual carp males were frozen in 0.5 ml straws by conventional freezing. Cooling rates were determined by monitoring the sample's internal temperature. We compared four freezing protocols, which involved placing sperm samples at various levels (1, 3, 6, and 9 cm) above the liquid nitrogen (LN) surface (corresponding to -190, -150, -110, and -70 °C, respectively) for 20 min followed by transferring the samples into LN. Freezing at 3 cm above the LN surface resulted in the highest motility (33 ? 8 %) and velocity (118 ? 9 ?m/s) of spermatozoa after thawing and diluting in swimming medium. We determined that -90 °C is an optimal temperature at which immersing the samples in LN does not affect sperm motility after thawing. The sperm motility of samples immersed in LN before or immediately after the crystallisation point (-16 °C) was 0 %. Motility of spermatozoa cryopreserved with or without a seeding procedure was not significantly different after thawing. Therefore, we hypothesise that supercooling the sample during the conventional freezing procedure is not the main damaging factor during carp spermatozoa cryopreservation.

The impact of non - functional sole of foot on the posture of people with non - sedentary kind of employment

This thesis focuses on the non-functional sole of the foot and its influence on people with sedentary job's body posture. As a non-functional sole of the foot is considered, in the research part of this thesis, the deviation of the standard that can be determined by means of podoscope which displays the sole of foot, or by functional tests. The first aim of this work is to describe the posture disorder of people working in an environment where they burden the sole of the foot in the long term. The second aim consists in the suggesting and applying a physiotherapeutic plan for particular people with a dysfunctional foot sole and not having sedentary job. The target of the research question was to define a particular pattern of the posture disorder as a consequence of the dysfunction of the foot sole. In the theoretical part of the thesis the reader is informed about the development and anatomy of the foot, its function that can be focused on from the point of view of a foot as such as well as the function of the foot as an impact on the posture when sitting, standing and walking. Another part deals with the posture and its modifications in both, the statistics and dynamics. In relation to the research part of this thesis, there are incorporated chapters about the possibilities for the examination of the foot from the point of inspection, palpation, active and passive movements and, last but not least, the functional examination including tests of the sole of the foot and instrumental examination of the posture - posturography. Because of the target of the description of the consequences of the non-functional foot sole, there is a description of the function muscle chains that connect the sole of the foot, and the posture, included. A short part of the theory is dedicated to sanitary footwear. For the research part of the work, three women aged 21 22 with non-sedentary jobs, working eight hours per day as a minimum, with subjective difficulties with the pain of lower limbs and back in the lumbar area, were chosen. The collection of data took place in the form of processing three case interpretations with the use of the method of an interview, comparison and a kinesiological analysis, including the use of objective methods of the examination with podoscope and posturograph. The content of the kinesiological analysis was focused on a chosen test that were after the end of the therapy registered in a chart depicting the pattern of the posture disorder. The examination by posturograph includes projection of the focal point of probands to the supportive base and the measure of their motoric abilities. There was a physiotherapeutical plan conducted and applied in the form of exercise during the therapy of probands. The therapy took place for nine weeks, the brobands exercised every two weeks under the supervision of a therapist with the purpose of eliminating potential mistakes. The exercise was extended or corrected according to the actual condition of probands. From the results of the theoretical part it is obvious that it is possible to create the pattern of the posture disorder in case of the people with non-sedentary working place.

Zdravotní průzkum dřevin lesoparku v intravilánu Hradce Kralove na soutoku Labe a Orlice

In the period from January 2013 to April 2014 was realized health survey of wood in the park Jiráskovy sady in Hradec Králové. In the first place survey was focused on occurrence of wood fungus. Introduction of the thesis is devoted to the localization, historical context, and then the characteristics of natural conditions and wider territorial relations. In the next section, is putting emphasis on inventory of affected wood and woody fungus, that points are the main cause of the investigation. These funguses were identified visually by fruiting bodies and describes their appearance, and damages that may result. There were determined mainly wood funguses like saprophytic character. Most funguses were found on individuals of Acer campestre and Aesculus hippocastanum.

The territory of south Bohemia Region from the point of the risk occurrence of insurance events

The purpose of this work is to analyze this territory from the point of the incidence of life-threatening and property-threatening incidents. The theoretical port contains all individually described kinds of insurance that eliminate the incurred losses. The practical part is reserved for specified and acknowledged risk of South Bohemia Region. Then possible arrangements are proposed leading to minimalizing of losses that may incur as a result of these risks. The practical part is closed by comparison of elected insurance products of four insurance companies. It concerns following sorts of insurance: insurance contents, insurance of buildings, insurance against damage in common civilian life, insurance against loss and damage resulting from the possession of buildings, the motor third party liability insurance and accident insurance. Every insurance company has its own general insurance conditions for its insurance products. In this part the differences in insurance conditions among the insurance companies are described. In conclusion the evaluation of gained results is presented.
