Kompozity nanouhlíkatých materiálů pro ochranu zemědělských rostlin před houbovými patogeny

Graphene oxide (GO) belongs to carbon-based nanomaterials (CBNs) and is capable of bonding with various molecules, thanks to the presence of functional oxygen groups in its structure, created during graphene oxidation. This mechanism can be applied on nutrient and pesticide delivery and improve their utilization by plants. Sulphur is an essential nutrient and also has fungicidal properties. In this study, graphene oxide was synthesized and used to make two nanocomposites containing sulphur. GO and nanocomposites were then characterized. Nanocomposites were tested for phytotoxicity, and their potential fungicidal properties were studied. Both nanocomposites showed significant fungistatic effect against grey mold when tested on plates with medium. However, when tomato leaves were treated with nanocomposites, inhibition effect against mycelium growth was ineffective.
Hobby akvaponie

The aim of this bachelor thesis was analysing hobby aquaponics systems available on the market and then designing, assembling and testing own aquaponics hobby system which can be used on balconies, terraces and gardens. Five different media beds and one combination of media bed and raft system were selected and evaluated. From the identified shortcoming of systems on the market were designed, assembled and tested in practice two media beds aquaponics systems. These systems were evaluated and compared with systems available on the market.
Rating the resistance of tomato (\kur{Lycopesicum annum }L.) cultivars to late blight (\kur{Phytophthora infestans})

The aim of this thesis was to evaluate resistent varieties of tomato in the term of susceptibility to late blight. Late blight , caused by P. infestans, is the most destructive foliar disease of tomato. Some new sorts declare the resistence to late blight. The aim of our thesis was to evaluate how these varieties perform in Czech republic. It was established experiment with 14 resistent and 2 control varieties. The leaves of the plants were examined routinely for symptoms of late blight and the severity of them was recorded. The varieties Mountain Magic, De Barao, Cherry Bomb, Tomato Des Comodores, Matts Wild Cherry a Plum Regal proved high level of resistence to late blight. The varieties Phantasia, Phytovia and Mountain Merit showed lower level of resistence. Growing new varieties, with resistence to late blight can enable the reduction of fungicidal protection of tomatoes and for organical and hobby growers to make possible growing tomatoes without use of pesticides.
Diagnostic method for Clavibacter michiganensis subsp. michiganensis, the causal agent of bacterial wilt of tomato

The aim of this methodology was optimize detection and determination of quarantine bacterium Clavibacter michiganensis subsp. michiganensis, the causal agent of bacterial canker of tomato, The disease causes high ecomomic losses in main production areas on indeterminate greenhouse and fi eld tomatoes. Methodology is based on a combination of both optimized techniques utilizing different principles of action: immunochemical (IF) and molecular (PCR). The submitted diagnostic protocol alows to eliminate potential false negatives and positives and ensure the highest reliability of results. Specific and timely detection of Clavibacter michiganensis subsp. michiganensis will alow effective aplication of plant protection mesures.
Isolation of antimicrobial peptides and proteins from tomato

Recent emerging problem is occurrence of multiresistant pathogens. This inconvenience makes people to search for new antimicrobials. One of possible ways is looking for natural antimicrobial peptides. In our project, green tissues of tomato (Solanum lycopersicum) were chosen for isolation of such peptides. Extracts obtained were fractionated by solid phase extraction and liquid chromatography. Antimicrobial activities were tested on model microorganisms. Several fractions exhibited antimicrobial activity against bacteria and fungi. In the extracts, precursors of two antifungal proteins were identified.
Pests of greenhouse cucumbers and tomatoes - informative and educational database

Diploma work is based on constructing of electronic informative system about greenhouse growns damaging pests - growtheal vegetables, concretely forced on forwarding cucumbers and tomatoes. The aim of diploma work is to make out the given information system with division of pests according to kind of the growth, place of damage and integrated locator of pests based on their Czech or Latin title, eventually based on information about pests and bioagens. The system contains detailed information about individual pests, their bionomii, enlargement and protection, including the biological protection. A part of this informative system is a large number of photographs of individual pests, biological preparations or injured plants with apparent symptoms.
