Changes in soil microbial community folowed using microbial fatty acids profiles

In this study the possibilities of microbial fatty acids profiles application for the integrated view on soil microbial community are discussed as well as the advantages and disadvantages of this approach.

Oxyggen consumption was used for measuring growth parameters of functional microbial groups in two soils of different texture after addition of mineral, sugar-ammonium and complex media. Phospholipid fatty acids analysis (PLFA) was used to determine differences in community composition and structure. Growth parameters and composition based on a complete PLFA pattern of the microbial community, changed as a response to medium addition and type of incubation. The proportion of actinomycetes decreased whereas the proportion of fungi increased after addition both of the sugar-ammonium and complex media. Gram-negative bacteria dominated after addition of the substrates rich carbon source, while Gram-positive bacteria prevailed after addition of the carbon and nitrogen rich medium.

This paper summarizes different approaches based on fatty acids analyses used in investigation of soil microbial community qualitative changes during primary succession on post mining sites (0-42 years). The evaluations of different community level were shown: (i) Community as a superorganisms based on complete phospholipid fatty acids (PLFA) profiles; (ii) Guilds of community based on specific PLFA biomarkers; (iii) PLFA pattern of cultivable community part; (iv) Bacterial strains level determined with help of cellular fatty acids methyl-esters (FAME, MIS Sherlock).
