Strukturní studie serpinu Iripin-5 z klíšťat \kur{Ixodes ricinus}

The aim of this bachelor thesis was a crystallization of serpin Iripin-5 from ticks Ixodes ricinus. Iripin-5 is the most produced serpin in the salivary glands of ticks. The focus of this thesis was to prepare protein Iripin-5 and perform crystallization experiments. Obtained crystals of Iripin-5 were in diffraction quality.

Optimal conditions for serpin Iripin-4 expression and purification were studied. Then, suitable conditions for protein crystal growth were investigated. These conditions were optimized and the formed crystals were tested.

Point mutation altering arginin for tryptophan amino acid residue in P1 site of tick salivary serpin Iripin-1 was created using specific primers. Recombinant protein with this mutation in nucleotide sequence was then expressed in chemically competent Escherichia coli cells, extracted from them and purified by affinity and size-exclusion chromatography. To see the impact of the mutation on inhibitory function of Iripin-1, its ability to bind trypsin and form covalent complexes was evaluated.

Antihemostatic effects of four recombinantly produced serpins of the tick Ixodes ricinus (IRS 2, IRS 3, IRS 5 and IRS 8) were tested using a method of flow cytometry. Fluorescently labeled antibodies against platelet surface molecules were used to determine the difference in platelet aggregation and activation after incubation of samples with specific serpin. To see how serpins affect coagulation pathway, the prothrombin time, activated partial thromboplastin time and thrombin time tests were performed.
Characterisation of novel serpin TILIr and its relatives from the superfamily of serine protease inhibitors from \kur{Ixodes ricinus} tick

Novel gene, named TILIr ( TIL Ixodes ricinus), that encoded yet unknown protein with TIL domain characteristic to proteins that belong to trypsin inhibitors family was discovered and isolated from hard tick Ixodes ricinus. Two isoforms of novel gene were identified and studied in this project. As the result of this study recombinant TILIr protein was produced in bacterial expression system and analysis of its functional characteristics was conducted. The protein belonging to family of trypsin inhibitors represent a potential candidate for anti-tick vaccine component.
