Study of the antioxidative potential of selected fruit marcs

Subject of this bachelor thesis was antioxidant activity of selected kind of fruits. Among selected kinds were the marc of red and white grapes, moldings from red elderberry and aronia. Two species of made capsules from marcs of white grapes and aronia were also subjected to analysis. Results of this bachelor thesis are summarized to four chapters. The first chapter compares the water content of drying at 40 ° C and 105 ° C. In the second chapter, the results of extraction in different proportions of solvent mixture ethanol - water and results of extraction in prepared artificial gastric juice are discussed. As a result, the water – ethanol mixture was a more effective solvent than artificial gastric juice. The third chapter deals with the results and discussion of the determination of the total content of phenolic substances. For the solvent mixture, the largest amount of polyphenols in the red vine marc was determined to be 43,56 mg.g-1, while in gastric juice the largest content in the elderberry was determined to be 11,89 mg.g-1. Capsules were also extracted in gastric juice, and 2,53 mg.g-1 was found in aronia tablets, while 5,93 mg.g-1 in white grape tablets. The last chapter compares the results of TAA measurements using the TEAC and EPR methods. The EPR method was more suitable for much diluted solutions than the TEAC method. The results of this work show that there is a large number of polyphenols and other antioxidants in the moldings that affect the overall antioxidant activity. Marcs have great potential for use as natural dyes or can be used to make a food supplement.
Foliar fertilizers containing collagen hydrolyzate for nutrition of grapevine: applied methodology with certification

A comprehensive methodology describing the use of products made from collagen hydrolyzate for leaf nutrition of main field crops and vines grown in conditions of organic farming as well as within the conventional method of cultivation. Elaboration of the methodology was preceded by applied research to develop environmentally acceptable ingredient - collagen hydrolyzate, where undesirable sodium is replaced by biogenic, often scarce potassium. Another objective was to optimize content and a mutual ratio of trace elements from the perspective of requirements of particular crops or their groups. Within the methodology are stated all the information needed for qualified decision-making regarding to foliar application of newly developed products in organic and conventional mode of farming. Examples of practical use of both types of fertilizers are the result of their application in field trials and South Moravian vineyards with standard and biological growing system.

A metodical approach for sanitation of grapevine cultivars using chemotherapy

The guideline describe a method of grapevine cultivation in vitro and its sanitation from virus pathogens usinf Ribavirin. Large part of this guideline describe diagnostic protocols for estimation of healthy status of sanitated grapevines. The guideline is intended for laboratories dealing with sanitation and diagnostics of viral pathogens of grapevine cultivars and rootstocks. Further utilization can be done by the Ministry of Agriculture, Central Institute for Supervising and Testing in Agriculture and also by growers of grapevine.

Comparison of content anthocyanin pigments in selected cultivars of black elderberry and grape

The purpose of this thesis is to compare the contents anthocyans in different varieties of European elderberry (Sambucus nigra L.). It was also studied the relationship between the content anthocyans in selected samples of blue grape varieties (Vitis vinifera L.) and position of the track, where the grapes were grown. The theoretical part describes elderberries and grapes in terms of botanical, chemical, and in terms of their using in the food industry. The dominant part of this chapter is to describe anthocyans and possibilities for their determination in the elderberry and grapes. Emphasis has been placed on the pH-differential method and determination by high performance liquid chromatography. The experiments are divided into two parts. The aim of the first experimental part is comparison of the total anthocyanins content in fifteen varieties of elderberry. The performed analysis shows that in terms of anthocyanins content for cultivation of elderberry in the Czech Republic the most suitable varieties are Mammut, Samyl, Reis aus Vossloch and Haschberg. In the second part of the experiments were determined total anthocyanins content in nine varieties of blue grape grown in the Mikulov region. For determination were used HPLC and pH-differential methods, as well as in the first part. The highest content of pigments was found in varieties Alibernet and Neronet. The present study focused on the suitability of the location pointed to vineyard south to southeast orientation plantings. At the end of the experimental part is a comparison of both methods of determination. Method using high performance liquid chromatography to provide very accurate results compared to less demanding pH-differential method of determination. This is recommended only for tentative determination of anthocyanins content in real samples.

Master´s thesis deal with optimum proposal reset vineyard economy in viticultural area Strachotin. The task is utiliziation of landscape potential CHKO Palava and hydro-electric plant Nove Mlyny with promotion development agricultural tourism in the area. Theoretic part of thesis includes analyses inquiries and progression areas, production problems and vine processing. Practical part includes set of questionable drawings, used technology and material for security of correct performance function construction.
The adjustment of the feed ration for laying hens in order to improve the strength and the hardness of eggshells

This major thesis expands the issue of the bachelor thesis "The issue of raising laying hens considering on introduction of enriched cages" and adds new knowledge relating to the topic feed rations and their influence on the final production. The theoretical part of the thesis analyses the basic theoretical concepts cohering with this topic, focused on creating a comprehensive overview of the structure and quality of the eggs, the basic ingredients in the feed rations, qualitative parameters of grapes as the modified additive provided by us and last but not least there is processed the recent overview of the current state of large-scale farmers in the Czech republic. The practical part of the thesis aims to test the effect of the proposed feed with an additive in the form of the crushed grape seeds, on the strength of the eggshell, by homogeneous samples of the breeds through from us proposed measuring instrument. Subsequently, a statistical evaluation of measured results is realized. In conclusion, the consideration is extended to the economic evaluation of costs and profits in case of the implementation of the suggested feed ration into practice in order to quantify the difference compared to the commonly used feed ration.
Influence of cultivation conditions relating to climatic change on the selected polyphenol content in grapevine plants

Winegrowing is highly climate sensitive. Even small changes in temperature could have big impacts on the whole industry. Because of the relatively high crop value, many growers can aff ord to invest in adaptation resources that might not be feasible for less valuable crops. Th ere are two basic types of response to climate change: working to adapt to or manage the impacts experienced, and trying to prevent or minimize further change (mitigation). Th ese are not mutually exclusive, and in fact, both are increasingly regarded as necessary. In our contribution we will discuss the eff ects of drought, irrigation and cultivation in greenhouses in comparison to cultivation in vineyards on the content of trans-piceid and trans-resveratrol in white and blue varieties of Vitis vinifera plants.
Cultivation of grapevine in conditions in vitro

Grapevine is a traditional crop in the Czech Republic, especially in the South Moravia region. It is a vegetatively propagated crop; hence vineyards are endangered by abiotic and biotic stress influence. The risk of plant damage due to frost and pest influence occurs in the field conditions according to season and locality. Cultivation of grapevine in conditions in vitro eliminates the risk. Higher utilization of the method is limited by sensitivity of some genotype to in vitro conditions. This methodology solves these problems and describes material introduction into in vitro conditions, explants multiplication and slow growth condition establishment. The Ministry of Agriculture of the Czech Republic is the user of this methodology and it will utilize it in the framework of “National Programme on Conservation and Utilization of Plant, Animal and Microbial Genetic Resources for Food and Agriculture”.

Grapevine cryoconservation

Storage of grapevine germplasm is endangered by biotic and abiotic stressors in the field conditions, and it is connected with the risk of valuable accession loss. Cryoconservation is a method which eliminates the risk and makes a safe long-term storage of plant germplasm possible. Cryoconservation method is based on storage of plant material at a temperature of liquid nitrogen when all processes are practically stopped but the viability of plant material remains. This methodology describes preparation of plant explants before cryoconservation, apical shoot tip dehydration by cryoprotectants, procedure and principles of long-time plant storage and plant recovery for evaluation of explant survival and regeneration. The Ministry of Agriculture of the Czech Republic is the user of this methodology and it will utilize it in the framework of “National Programme on Conservation and Utilization of Plant, Animal and Microbial Genetic Resources for Food and Agriculture”.

A metodical approach to detect viruses belonging to a genus Vitivirus infecting grapevines in the Czech Republic

A metodical approach describes detailed procedure to detect two important grapevine viruses belonging to a genus Vitivirus. Two independent methods are recommended for the detection: RT-PCR and DAS-ELISA.
