Role of adenosine deaminase in regulation of energy during bacterial infection in adult Drosophila melanogaster

The expression of adenosine deaminase related growth factor A (ADGF-A) during immune response was studied in Drosophila melanogaster. The effect of increased extracellular adenosine signaling due to hemocytes specific ADGF-A knockdown was investigated by subjecting the flies to the chill coma experiments.

The aim of this thesis was to measure adenosine production from lymph glands in D. melanogaster larvae ex vivo and to assess the differences upon infection with parasitic wasps L. boulardi. The second aim was to investigate the influence of two different nucleotidases on adenosine levels upon gene knockdown of their respective genes with and without a triggered immune response.

Extracellular adenosine triphosphate and extracellular adenosine are important regulatory molecules in the human immune system. The concentrations of these molecules are in turn regulated by ectonucleotidases and adenosine deaminases. In this thesis I attempt to test the gene silencing efficiency of RNA interference for three different genes coding for such enzymes in the model organism Drosophila melanogaster.
Genetic screening for the dominant suppressors of adgf-a mutant phenotype in Drosophila

Extracellular adenosine is an important signaling molecule in various physiological processes. When not regulated properly it can cause various pathologies such as Severe Combined Immunodeficiency. To study the effects of increased extracellular adenosine level a Drosophila model has been established. This model is based on mutation in the main adenosine deaminase gene in Drosophila (ADGF-A, Adenosine Deaminase-related Growth Factor A). This work is a consequence of large screening for the suppressors of adgf-a mutant phenotype and presents a more detailed study of 3 previously identified regions containing dominant suppressors.
