Hybridisation of cyprinid fishes in the Czech Republic: genetic and morphometric analysis of the subfamily Leuciscinae

Hybridization is a process when two interspecific individuals mate and spawn. When backcrossing it often goes hand in hand with admixing unrelated genome into the genome of a species. Nowadays it is well established that hybridization is not as uncommon as it was believed. Especially in vertebrates fish are quite common in such phenomena. In Leuciscids, the family of order Cypriniformes is relatively frequent. It is a rife family in the European freshwater systems. Factors that contribute to such frequent hybridization are extraneous fertilization, sympatric occurrence, competition for spawning grounds, and other ecophysiological traits. It was known since the second half of 20th century that this is an ongoing phenomenon in many species of this family. Previous research was focused mainly on morphological traits. The relative pinnacle of such research was based on allozymic analysis. The goal of our project was to evaluate the level of hybridization in the main water bodies of the Czech Republic with the main focus on the detection of involved hybrid species, the level of such hybridization, and the phenotypic characterization of hybrid individuals. Evolution of visual sensory system in the common barbel (Barbus barbus)

Vertebrates inherited five visual opsin classes from their common ancestor expressed in the rod and cone cells. While many vertebrate groups reduced the number of visual pigments they possess, opsins of teleost fishes have diversified. Today, teleosts outstand among all vertebrates in the number of visual pigments and photoreceptor types. This extraordinary development was also facilitated by the teleost- specific whole genome duplication that occurred approximately 350 Mya. Here I explored teleost species that recently underwent subsequent whole genome duplication - the common barbel (Barbus barbus). This event was accompanied by hybridization (i.e., it was allopolyploidization) and resulted in 14 opsin genes found in the genome, an unusually high number even among teleosts. In this thesis, I studied the effects of such an expanded opsin repertoire on the visual abilities of the common barbel. Based on the whole genome, I have completed the reference sequences of all opsin genes and revealed their positions on the chromosomes. Fish community trophic structure in antrhopogenically perturbed rivers

Využití stabilních izotopů pro hodnocení trofické struktury společenstev nabývá v posledních dekádách na popularitě a stává se tak stále více používaným nástrojem užívaným k výzkumu trofických pozic a potravních nik. V této práci jsou shrnuty základní principy, které se při práci se stabilními izotopy využívají, představeny mechanismy výměny tkání, izotopového routingu a diskriminačního faktoru, jejichž zanedbání může vést až ke znehodnocení nabytých poznatků. Dále jsou v práci představeny základní metody využití stabilních izotopů, jejich výhody a nevýhody, matematické modely zpracování dat a jejich uvedení do praxe pomocí praktické části, ve kterém bylo zkoumáno trofické postavení a niky kaprovitých ryb v antropogenně ovlivněných řekách.

The evolution of vision in ray-finned fishes

Vision plays a key role in life of many vertebrates, and the performance of visual system is often adapted to specific environments inhabited by individual species. Fish colonized a wide range of habitats and adjusted their visual abilities to maximize their success rates in hunting, reproduction and predator avoidance. This thesis is focused on molecular mechanism of visual system, namely on genes for photoreceptor proteins, opsins, of two major groups of teleost fishes: African riverine cichlids (family Cichlidae, order Cichliformes, part of larger taxa Percomorpha), and European freshwater cyprinids (family Cyprinidae, order Cypriniformes, part of larger taxa Ostariophysi). Two types of photoreceptor cells are present on retina: the cones and the rods. Actinopterygian fishes in general have four cone opsin types (SWS1, SWS2, RH2 and LWS) used for colour (photopic) vision, and one rod opsin type (rhodopsins) for vision in deteriorated light conditions (scotopic vision). In the present thesis, I focus on 1) DNA sequence and amino acid substitutions of the opsin genes, and on 2) gene expression levels of opsins sensitive to various wavelengths of light spectrum. Littoral age 0+ fish distribution in relation to multi-scale spatial heterogeneity of a deep-valley reservoir

The distribution of the littoral age 0+ fish assemblage was studied with respect to spatial heterogeneity of a deep-valley reservoir over the entire growing season. The abundance and diversity of age 0+ fish were also examined in different habitat types along the longitudinal profile of the reservoir during the daytime.

Patterns in 0+ fish distribution along longitudinal and depth profiles were studied in five canyon-shaped reservoirs in the Czech Republic. In two canyon-shaped and one shallow well-mixed reservoirs the efficiency of fry trawls was investigated during late summer for juveniles and in one canyon-shaped reservoir the efficiency of ichthyoplankton trawls was investigated during spring for larvae and early juveniles.
