83 Abstract The work focuses on the physiology of wood-rotting fungi and the possibilities of their inhibition by physical methods (microwave and gamma irradiations, dry heat) or newly developed chemical preparatives contaning metallic nanoparticles. The effect of abovementioned treatments on mycelial growth, activities of extracellular enzymes responsible for wood degradation and on spore germination or viability was studied in selected strains of wood-rotting fungi obtained from culture collection (Coniophora puteana, Fibroporia vaillantii, Gloeophyllum sepiarium) or isolated from infected buildings (Serpula lacrymans).

Physiology of Serpula lacrymans

Serpula lacrymans, the causative agent of dry rot indoor wood decay, is the most common Basidiomycete found in buildings. From the viewpoint of economic loss and destructiveness it is the one of the most important fungus worldwide. S. lacrymans is able to fastly colonize substrate by rapid mycelial spread and effectively decompose it. Despite the fact, that the physiology of this fungus has been studied for more then 100 years, efficient approach for efective defense does not exist. However, the prevention and effort to keep the house enviroment dry still seems to be the best option. This thesis is a survey of a recent knowledge of S. lacrymans physiology and up-to-date technologies used in the remediation of affected buildings. key words: Serpula lacrymans, fungal physiology, rhizomorphs, remediation, wood construction, hot air, microwaves, fungicides

The work was focused on the current state of the object with high humidity from the ground in presence of fungi. The aim was to consider possible ways to restoration of it and to elaborate the final version of the restoration proposal. The semi-destructive methods in the construction survey were used. A humidity of endangered construction elements was measured, the degree of infestation and its intensity were determined. Results were entered into drawings supplemented by tables. Probable cause of damage and possible ways to eliminate it were in results also recorded. Such a combination of options to ensure the long-term solution to the problem with regard to maintaining the authentic appearance of the building by creating benchmarks and comparison of the results was chosen. This combination includes a drainage system, an insulation of lower structure against moisture and creating a vertical drainage, ensuring the floor and the interior ventilation with the help of horizontal openings in the foundation wall, roof structure repair, replacement of the roofing and ensuring stiffness of the truss. The results of this work provides the necessary documents to the contracting authority to take the necessary steps in the reconstruction and rehabilitation of timber structures, respectively, to award a professional company. It can also be applied to objects of a similar nature that are also threatened by high incidence of moisture from the lower building.
