The use of near-infrared spectroscopy for characterisation of raw material for sunflower oil production

This diploma thesis provides summary information about using of near infrared spectroscopy for monitoring of high oleic acid content in sunflower seed. The calibration for high oleic sunflower seed was performed by using calibration model PLS - „Partial least squares. Gas chromatography was used as a reference method. Based on this knowledge thirty samples of high oleic sunflower seeds were measured and evaluated on this calibration. The last part of diploma thesis consist of comparison of oleic acid results measured by near infrared spectroscopy and gas chromatography used as reference method, and discussion about the measurement accuracy and evaluation of suitability of using these methods.

Usefulness for discrimination caffeine and decaffeinated coffee is tested. Next, flour samples are researched. There is examination of presence or absence of gluten.

The aim of this work was to create an overview of the possibilities of using NIR spectroscopy for evaluation of grain quality and evaluation of the influence of malting barley varieties, test site and a treatment system for the crude protein content of grain (hereinafter referred to as NL) from harvest 2018.The NL content was determined by NIR spectroscopy. It was found that the change in NL content was statistically significantly influenced by varieties and habitat. The NL content ranged from 12.2 to 13.9 % as average in all test areas. Favorable NL content was found in the Cereal Testing Area (11.9 %). There were statistically significant differences between the treated and untreated variants, except for the Beetroot area. In all test areas, the NL content was affected by a habitat of 79-87 % and a variety of 4-10 %. In the corn test area, the change content is due to 10% variety and 79 % to habitat. On the contrary, in the potato testing area, the variety's influence is only 4 % and the habitat influence is 87 %. The possibility of using the NIR spectroscopy method for barley quality assessment in the case of NL in grain was successfully verified.

The dissertation thesis is focused on investigation of possibilities of usage NIR spectroscopy for nutritional composition of fruit and vegetables evaluation. The basic nutritional components of fruit and vegetables, methods of apples, peaches and tomatoes storage are characterized in the literary section. The history of infrared spectroscopy, present, principles and types of NIR spectroscopy analyses are described in detail. The possibilities of usage of NIR spectroscopy in food industry aimed on fruit and vegetables quality evaluation are also described in the literary section. Changes of fruit and vegetables nutritional composition during ripening are evaluated in the experimental part. Evaluated fruit and vegetables were apples (variety Rubinola), tomatoes (variety Dagmar) and peaches (variety Redhaven). Changes in the total polyphenols content, titratable acids, soluble dry matter, malic acid, ascorbic acid, citric acid, peel firmness and apple flesh firmness were evaluated. Calibration models were prepared from the gained data using the NIR spectroscopy method. The changes of stored fruits nutritional composition were evaluated using the created calibration models. Soluble dry matter, titratable acids, ascorbic acid and flesh firmness of different apple, peach and tomato varieties can be determined fast and exactly according to the data. The accuracy of models for total polyphenols and peel firmness is significantly influenced by the number of calibration standards. The accuracy of models for malic and citric acids is influenced by the concentration of these components in fruits.

Current trends in analytical chemistry is the introduction of instrumental methods that are quick and economical. This process is obviously not avoid potato field of chemistry, using optical methods - NIR spectroscopy. This thesis therefore focuses on the analysis of basic nutrition and most of all the determined parameters of potato tubers (dry matter, starch, N-substances and reducing sugars) using laboratory reference methods and their subsequent transfer to this new analytical method by creating appropriate calibration models. It is also part of the work is devoted to the statistical comparison of both analytical methods in order to determine whether these methods can replace each other. The measurement has been found that some parameters NIR spectroscopy can be replaced, while in others the conversion is not suitable. Specifically parameters starch, N-substances or dry matter had correlation coefficients (R) 0.901; 0.806 and 0.977, meaning satisfactory calibration models. Whereas on parameters carbohydrates, carbohydrates after hydrolysis with the values of correlation coefficients 0.769 and 0.786 is impossible to apply the method of NIR spectroscopy. Models for determining these are not suitable.
