This diploma thesis deals with the design of a hydrodynamic seal of an axial pump with an annular motor, and it does so in three main thematic planes: research of current knowledge, simulation and experimental. The research section provides an overview of a broad problem of magnetic fluid seals (ferrohydrodynamic (FHD) seals), current developments in the field of simulation of internal flow in magnetic fluid and in the field of application of FHD technology in sealing liquids. It also covers a wider context of potential use for mechanical circulatory support systems that have had a significant impact on the proposed concept of a novel axial FHD seal. The remaining two thematic planes deal with his design and experimental testing.
The flow of magnetic liquid with Bingham model application

Main topic of this thesis are magnetic fluids. These are specific type of fluids which can simplistically be considered as Bingham fluids. Main issues regarding the magnetic fluids mentioned in this thesis are: rheological properties of the magnetic fluids, behaviour of the magnetic fluids and the use of the magnetic fluids in industrial applications. Main goal is to apply Binghams model on the the magnetic fluids assuming that this model can be applied with a good accuracy. Equations which describe behaviour of the Bingham fluids are constructed. Some assumptions which have to be respected to use this analytical equations for magnetic fluids are formulated. Flow of bingham fluid is analytically solved in some simplificated cases with consideration of laminar flow. Analytical results are confronted with numerical ones obtained from CFD software Fluent for the purpose of verification.
How magnetic Fluids Affect the Germination and Initial Growth of White Mustard (Leucosinapis alba (L.) spach)

Text přináší výsledky z testování toxicity tří typů vodních magnetických kapalin (kyselý pH 2,7 citrátový pH 6,4 a alkalický ferrofluid pH 11,8) při koncentracích 0,10, ,25, 0,5 a 1,00mg nanočástic magnetických oxidů železa v 1 ml příslušné nosné kapaliny (surfaktant a rozpouštědlo). Sledovány byly klíčivost a základní charakteristiky počátečního růstu u namořených semen bělohořčice bílé. Významná toxicita byla zajištěna u kyselého ferrofluidu, naopak neurální vliv byl zaznamenán u citrátového ferrofluidu. Hodnoty sledovaných charakteristik a alkalického ferrofluidu přinesly zajímavá zjištění. Vyšší koncentrace byly toxické (0,5 a 1,00 mg/ml) a nižší koncentrace oxidů železa (0,10 a 0,25 mg/ml) částečně odbouraly negativní vliv účinku samotného nosného média (terametylamonium hydroxid).

Magnetic modification of microbial cells

Baker´s yeast (Saccharomyces cerevisiae) were magnetically modified by three different methods, namely, surface modification by magnetic fluid, entrapment of cells into alginate and covalent immobilization on particles of magnetic chitosan. The ability of H2O2 decomposition was tested for all types of modification. It is apparent that the most amount of hydrogen peroxid was degraded by magnetic fluid - modified cells (84-95%), while the efficiency of cell which were modified by other methods was much lower (40-60%). Thanks to immobilization on particles of magnetic chitosan, we made completely new type of magnetic material, which was tested for adsorption of Crystal violet and Safranin O. It was founded that magnetic chitosan adsorbs no dyes, so all adsorption belongs to immobilized yeast. The maximum adsorption capacities were determined using Langmuire isotherm at 69,4 mg/g for Crystal violet and 99,0 mg/g for Safranin O.
