Silver based raw materials for cosmetic purposes

The purpose of this bachelor’s thesis was a research about silver based cosmetic products and determination of antimicrobial activity on certain species of microorganisms by chosen silver based cosmetic products. Theoretical part is focused on the problematics of silver based cosmetic products, on their main purpose, physically chemical characteristics and toxicological profile. In the theoretical part is also mentioned problematics of legislative, mainly the safety and real effectivity of tested cosmetic products. Experimental part was focused on determination of antimicrobial activity of chosen silver based cosmetic products. Two silver based cosmetic product were used to determined antimicrobial activity on selected microorganisms Bacillus subtilis, Bacillus cereus, Micrococcus luteus, Kocuria rosea, Serratia marcescens a Candida vini. From result it is apparent, that both of used Cosmetic product showed antimicrobial activity, but not on every used microorganism.
Development and preparation of antimcrobial nanostructure biomaterials

The presented diploma thesis deals with the optimalisation of preparation and the preparation of combined nanostructured antimicrobial biomaterials itself. In the theoretical part, a review focused on used materials and consequently preparation of nanoparticles and nanofibers was elaborated. Furthermore, the used antimicrobial substances – curcumin and ampicillin, and the principle of cytotoxicity assay were described.In practical part the optimalisation process is described. Furthermore the safety of prepared materials and used antimicrobial substances on HaCaT cell line was tested, in order to confirm their possible further use in cosmetic and pharmaceutical industry. Great part of the thesis deals with evaluation of the antimicrobial activity of used substances and prepared combined nanomaterials on multiple microorganisms from grampositive bacteria, gramnegative bacteria and yeasts. Also the release speed of antimicrobial substances from prepared nanomaterials was determined by spectrophotometer. The amount of released ampicillin from prepared nanomaterials was determined by liquid chromatography.
Use of cell cultures to testing of natural substances of plant origin

The presented bachelor thesis is focused on preparing aqueous and oil extracts of plant origin, their characterization and testing of their cytotoxicity and genotoxicity. The theoretical part contains basic information about plants and kinds of spices or tea that were used. Description of used methods of liposome preparation and encapsulation of active substances, measurement of antioxidant activity, particle characterization, cultivation of cell cultures and the principle of cytotoxicity and genotoxicity tests used in this thesis is included. Phenolics and flavonoids content was determined using the spectrophotometry. Antioxidant activity was evaluated spectrophotometrically as well. By ultrasonic homogenization method, liposomes were prepared and active agents were encapsulated into these vesicles. Encapsulation efficiency and stability of active substances were measured by spectrophotometry. By DLS method, size of particles was determined. Particle stability was derived from the zeta potential value. Important part of the bachelor thesis was cultivation of human keratinocytes. The cytotoxicity of extracts and liposomes was assessed by MTT assay on human cell cultures. The genotoxicity of extracts and liposomes was defined using SOS Chromotest on model organism, genetically modified E. coli. In conclusion, the potential use of prepared liposomes in the cosmetic or pharmaceutical industry has been proposed.
Development of cosmetic products with antimicrobial effect

The proposed bachelor's thesis focuses on the preparation and study of nanomaterials with antimicrobial properties. The nanofibers were synthesized from biopolymers using the forcespinning method, and liposomes were prepared using the sonification method. Active ingredients used in this work are tymol, carvacrol, and eugenol. The theoretical section of the thesis discusses the methods for preparing nanofibers and nanoparticles and the materials required for their synthesis. It also covers the encapsulation of active ingredients in nanomaterials. Additionally, this part explores the skin microbiome, antimicrobial effects, and the use of both natural and synthetic antimicrobial agents. The practical part of the thesis focuses on determining the polyphenol content and antioxidant activity of each active ingredient. The gradual release of active ingredients into saline solution and distilled water was determined for nanofibers synthesized from polyhydroxybutyrate with encapsulated active ingredients. Furthermore, it deals with the optimization of nanofiber synthesis from chitosan and polyamide using the forcespinning method. The functionalized liposomes were characterized in terms of their size, stability, encapsulation efficiency, and gradual release of active ingredients. The liposomes showed high stability even after several weeks, and their size ranged from 170.90 ± 1.63 nm to 243.07 ± 7.83 nm. Finally, the antimicrobial activity of the prepared products and active ingredients was tested against Cutibacterium acnes, Staphylococcus epidermidis, and Escherichia coli. The tested active ingredients in pure and encapsulated form, demonstrated antimicrobial effect against both gram-positive and gram-negative bacteria. We assume the possibility of using the prepared nanomaterials in cosmetic products intended for the treatment of acne and other skin diseases, and also its use in the form of preservatives.

Detection meticilin resistant strains of Staphylococcus aureus in material microbiology laboratory Laboma České Budějovice

The subject of my thesis is the detection meticilin resistant strains of staphylococcus aureus in the material of the Laboma Microbiology Laboratory in České Budějovice. This thesis is divided into a theoretical part and a practical part. In the practical part, the methodology of the detection and the identification of the methicillin resistant Staphylococcus aureus (MRSA) is specified in detail, as well as the exact procedures and the principles of the particular methods that were used. Further, there is the graphically and statistically processed data about the MRSA virus that was obtained in 2008 and 2009. The aim of this thesis was to determine the incidence of MRSA in outpatients. The comparison of the incidence of MRSA in both sexes ( that is a comparison of the individual months and comparison of the years 2008 and 2009). Furthermore, there was also the detection of resistance and sensitivity to the tested antibiotics and the identification of high-risk age groups. The result was that MRSA occurs most in the age group of those who are between 76-90 years of age, as surveyed in both years.The greatest resistance has been observed in the use of oxacillin and the smallest amount of resisitance has been observed in the use of Gentamicin. The largest number of positive isolates of MRSA were seen in February of 2008 and November of 2009.
