"DNA" barcoding is of limited value for identifying adelgids (Hemiptera: Adelgidae) but supports traditional morphological taxonomy

The presented study deals with adelgids (Hemiptera: Adelgidae) identification based on the sequence divergence of part of mitochondrial cytochrome ?c oxidase I (COI) gene used for ?DNA barcoding?. Analysis evaluates the DNA barcoding ability to discriminate adelgids on the genera level and it supports species identification based on morphological taxonomy. However, it failed to recognize species within species complexes.

Five molecular markers (mitochondrial AT-control region and COI, nuclear ITS and markers from primary enosymbiont Buchnera sp. argS and rrs) were tested for use in species identification of 2 selected genuses of the Adelgidae family. Markers COI, argS and rrs were succesfuly amplified and sequenced. Aligned sequences of those three markers were used for reconstruction of dendrogram by Neighbour-Joining metod with Kimura 2-Parameter model. These markers are suitable for resolution of taxonomic relationship of genuses within family Adelgidae, although further testing of these markers in conjunction with other markers (like ITS or AT-control region) will be required to delineate sister species with higher precision.
Optimization of cytogenetic techniques in a model aphid, \kur{Diuraphis noxia} (Aphididae)

Several cytogenetical methods for investigation of karyotyp were used in an aphid species, Diuraphis noxia (Hemiptera: Aphididae). These methods (FISH with telomeric probe, FISH with rDNA probe, FISH with histone probe and silver staining) were optimized for application to a sister group Adelgidae, where they could be useful for better differentiation of species in particular stages of their complicated life cycle.

Three molecular markers were tested for use in species identification of two selected genuses of the Adelgidae. Two mitochondrial markers were successfully amplified and sequenced, part of the COI gene and fragment between the COII and COIII genes. Their ability to distinguish species complexes was checked by statistical methods and by construction of phylogenetic trees.

Four molecular markers (nuclear ITS, EF1 alpha and mitochondrial AT-Rich and COI) were tested for use in species identification of Adelgidae family. Markers EF1 alpha and COI were successfuly amplified and sequenced. Aligned sequences of those two markers were used for reconstruction of dendrogram by Minimum Evolution method with Kimura 2-Parameter model. Comparison of dendrogram with assumptions based on morfology and biology of Adelgids confirmed suitability of chosen molecular markers for reliable resolution of taxonomic relationship within Adelgidae, although further testing and markers will be required to delineate sister species with higher precision.
