5 Abstract Cristina Stanciová Evaluation of activity of potentional antibacterial substances through the use of microdilution broth method II Diploma thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Specialist in laboratory methods The aim of this thesis was to test substances produced by the Department of Pharmaceutical Chemistry and Drug Control and the Department of Inorganic and Organic Chemistry Faculty of Pharmacy of Charles University in Hradec Králové. Substances were tested by using microdilution broth method. Testing was carried out on eight strains of the bacteria: Staphylococcus aureus, Staphylococcus aureus methicilin resistant, Staphylococcus epidermidis, Enterococcus sp., Escherichia coli, Klebsiella pneumoniae and Pseudoonas aeruginosa. Overall were tested 97 substances, which were divided into ten groups according to their chemical structure to: derivatives of (Z)-5- arylmethyliden-2-thioxothiazolidin- 4-on, anilids of pyrazinocarboxyl acid, arylaminopyrazins, pyrazin-2,3-dikarbonitrils, esters of pyrazincarboxyl acid, N- (2-anilino-1-alkyl-2-oxo-ethyl)-2-hydroxy-benzamides, esters of cholesterol and alkan΄s acids, salicylthioamids, styryl benzoxazols and benzoxazepin-dions. Statistical processing of the Hepatitis C virus genotype analysis results. Material and methods: In the period of 1 July 2005 - 30 June 2012, the Hepatitis C virus genotype analysis was performed in respect of 2 185 samples at the division of molecular biological diagnostics of the serology laboratory of the Institute of Clinical Biochemistry and Laboratory Diagnostics of the General Faculty Hospital and the First Faculty of Medicine of Charles University in Prague, using the reverse hybridization method. The testing results were statistically processed. Results: Of the set of 2 185 samples, 1 372 (62,8 %) originated from males and 813 (37,2 %) from females, with the average age of patients included in the set being 36,7 years. The majority of requests for the HCV genotype analysis originated from specialists in Prague (91,0 %). For drug testing was used the microdilution broth method. Author: Martina Drábková Title: In situ analyses of inflammatory colitis in Muc2-/- mice Diploma thesis Charles University in Prague, Faculty of pharmacy in Hradec Králové Pharmacy Background: Muc2 mucin is the main structural component of the intestinal mucus which forms important protective barrier avoiding the direct contact between the commensal bacteria and the epithelium. In order to understand the role of Muc2 mucin a Muc2 deficient mice strain has been developed. Initial observation showed that these mice already in few weeks after the birth develop spontaneous colitis and later on colon cancer. To better understanding what is going on in the colon and small intestine of Muc2-/- mice, in this thesis we focused on in situ analyses of distribution of different immune cells in these tissues. Methods: In our experiments three Muc2-/- , two Muc2+/- and three wild type mice on the C57BL/6 background were used. For the detection of distribution of different immune cells within lamina propria of small intestine and colon the immunohistochemistry analyses on sections from these tissues were performed. Lucie Křivčíková Title of the bachelor thesis: Candida species This bachelor thesis deals with a laboratory diagnosis of Candida spp. The main goal is to create a compact survey of laboratory methods which are utilised for a laboratory diagnosis of Candida species especially in the microbiological laboratory. The thesis is divided into four parts. The first part focuses on the common characteristic of the kingdom Fungi and the yeasts. The second part serves as the basic laboratory methods survey for a laboratory diagnosis of Candida spp. The primary classification of mycotic diseases with their basic characterisation is processed in the third part. Furthermore, this part includes the most frequent agents of these diseases from the genus Candida as well and the ways of clinical material taking. The fourth part is devoted to the characterisation of the most frequent Candida spp. agents of the yeast infections. Dvořáková Iveta Title: Human papillomavirus vaccination Diploma thesis Univerzita Karlova v Praze, Farmaceutická fakulta v Hradci Králové Field study: Specialist in laboratory methods Background: The aim of this work is to bring general awareness of cervical cancer, its creator, transmission, symptoms and treatment. It presents information about registered vaccines on the market. The aim of this study is to determine women and girls awareness of vaccination against cervical cancer, and their willingness to be vaccinated. Determine view - gynecologist vaccination against cervical cancer. Main findings: Vaccination against cervical cancer is already included in the vaccination calendar. Not an obligatory vaccination but only as recommended vaccinations for girls between 13 and 14 years. Conclusion: The diploma paper indicates the general awareness of the causative agent, transfer options and the prevention of cervical cancer among girls. The study found that most doctors inform girls and women about the possibility to be vaccinated. Doctors would recommend vaccination for men too. Most mothers are trying to have regular preventive examinations. The study found that most mothers are going to vaccinate their daughters, but they would not get vaccinated. Under certain circumstances, such as damage or injury of skin, mucous membranes and in immuno-incompetent individuals, the MRSA infection can originate. Therefore, hospitalized patients are much more susceptible to this infection. The biggest dangers represent MRSA (methicillin-resistant Staphylococcus aureus) strains. These strains of Staphylococcus aureus are mostly multi-resistant, where in addition to resistance to oxacillin (methicillin), resistence to other antibiotik occurs as well. Treatment of these patients is problematic, because MRSA strains are extensively resistant and persist in the organism and in the environment. Their eradication is not simple.
