The anthracycline antibiotic doxorubicin is widely used in the treatment of solid and haematologic malignancies, including breast cancer. The major metabolite of doxorubicin is 13-doxorubicinol that has a significantly lower antineoplastic potency compared to the parent drug and has been implicated as a potential mediator of chronic cardiotoxicity. The aim of the present study was to evaluate the effect of inhibitors of carbonyl reductase 1 (CBR1), such as quercitrin, rutin, and menadion, and prospective anticancer agent oracin on reduction and toxicity of doxorubicin in human breast cancer MCF-7 cells. Experiment performed with menadion revealed that CBR1 plays a important role in reduction of doxorubicin in cytosol of MCF-7 cells. Martina Gavelová, Ph.D. Title of a diploma work: Influence of selected cytostatic drugs on activity of cytosolic carbonyl reducing enzymes in MCF-7 cell line A major problem in cytostatic treatment of malignant tumors is the development of drug-resistance. One of the potential mechanisms of resistance is induction of drug-inactivating enzymes. The aim of the present study was to evaluate the effects of short-term exposure of MCF-7 cells to doxorubicin and oracin on the activities and expression of selected carbonyl reducing enzymes. Carbonyl reduction of these cytostatic drugs leads to 13-doxorubicinol (doxol) and 11-dihydrooracin (DHO), the major metabolites with lower antineoplastic potency compared to the parent drugs. We found that short-term (48 h) exposure of MCF-7 cells to low (nM) concentrations of doxorubicin or oracin caused a significant elevation of both cytostatics reduction rates. The diploma thesis deals with the issue of school readiness and its characteristics. In the theoretical part of the paper the problems of the appropriateness of school attendance postponement and the child?s school attendance preparation are worked up. Likewise the issue of children´s development of pre-school stage is treated and, not least, the diagnostic methods of considering the child school readiness methods and various tests used in practice including their evaluation are mentioned. In the practical part, the research is carried out based upon the observation, indigenous screening elaboration and the final evaluation.
