Optimization of chromatographic separation for the determination of 4-aminophenol and active substances in Valetol

This thesis deals with the optimization of separation conditions determine paracetamol, caffeine, propifenazone and 4-aminophenol in Proprietary Medicinal Products Valetol. As an analytical method was chosen HPLC with UV detection mainly for the following reasons: many other tablets contain excipients which are from the sample during analysis impossible, and separated themselves determination of active substances to monitor the stability of active substances The work was conceived as original, and the emphasis is on the greatest possible simplicity and less time consuming and optimized separation while maintaining high accuracy and reliability of the method. Optimal conditions suitable for the final validation of the separation were found on a column of C-18 RP-amide. Total analysis time does not exceed 13 minutes with equilibration after gradient elution of the column to its original condition. All substances are separated with a resolution greater than 1.5. The peaks of substances are symmetric and do not show significant peak tailing besides AMF. The zero line is despite the gradient elution stable.
Chemiluminescence determination of propylgallate using the flow injection analysis

7 Summary  The literature was searched for articles dealling with various ways of determining propylgallate and articles concerning the possible use of the FIA method with chemiluminescent detection in pharmaceutical analysis.  Measurement conditions were optimized. Appropriate device parameters, time and flow rate were set.  The optimum composition of the carrier stream was found and the influence of some substances to enhance chemiluminescence was tested. The effect on increasing the intensity of CL was detected for sodium polyphosphate, which increased 2.3-fold CL intensity.  Under optimum conditions the calibration curve was measured without addition of PFS and the results were evaluated.  Measurement conditions have been modified and calibration curve with the addition of PFS was evaluated.  Using the calibration curve with the addition of PFS the substance of propylgallate (Aldrich Company) and pharmaceutical formulation were analyzed.  Comparative method was chosen - Chelatometric titration. Also, using this method the substance propylgallate (Aldrich company) and formulation were assayed.  The results obtained by FIA with chemiluminescent detection and chelatometric titration were statistically evaluated using Student's test and Moore's t test and thus their comparability was demostrated.

PharmDr. Hana Sklenářová, Ph.D. Title of Thesis: Determination of fluoride in toothpaste using SIA system Determination of fluorides was tested in the toothpastes using the SIA system. For determination of fluorides reaction of aluminium ions with aluminon that forms a red complex was used. After addition of a fluoride solution, aluminium ions were released from this complex. Spectrophotometry was used for the detection. The calibration range for sodium fluoride was linear over the range of 3x10-3 to 1x10-2 mol.l-1 . The repeatabitily of the measurement was tested for blank solution (water) and for the solutions of sodium fluoride 5x10-3 mol.l-1 and 6.25x10-3 mol.l-1 . The repeatability was in the range of 1.19 - 3.70 %, that was found acceptable. Then, potencially interfering ions were identified, which may effect determination of fluorides. Following the composition of usual toothpastes interferents could be: Cl- , Br- , I- , Ca2+ , PO4 3- , Zn2+ , CO3 2- , Al3+ , CH3COO- , Sn2+ , tartrate anion, citrate anion, lauryl sulphate anion, sorbitol and methylparaben. Furthermore, attention is focused on the subsidy received from the ROP southeast. In summary is given a new programming framework 2014-2020. The practical part of the thesis was to describe, analyze and evaluate the project co-financed by EU structural funds. The project was implemented within the ROP SE and tackle integrated transport in the region Breclav.
