K experimentům byly použity vakuoly izolované ze suspenzních kultur šišáku bajkalského (Scutellaria baicalensis, Lamiaceae). Po provedení všech experimentů bylo zjištěno, že transport flavonoidů je nejvíce ovlivněn kyselinou glukuronovou v kombinaci s baicaleinem a MgATP. V přítomnosti kyseliny glukuronové a baicaleinu dochází pomocí endomembránových proteinů k syntéze baicalinu a k jeho ukládání do vakuol. MgATP tento proces ještě více zintenzivňuje a urychluje. In vitro cultures of medicinal of plants - VII Elicitation is one of the methods that can be used to increase production or accumulation of secondary metabolites in explants cultures. There were used callus and suspensions cultures of Silybum marianum (L.) Gaertn. Derivate N-phenylpyrazin-2-carboxamid was used as abiotic elicitor in concentrations c1=1.159x10-3 mol/l, c2=1.159x10-4 mol/l and c3=1.159x10- 5 mol/l. The effect of elicitors on the production (flavonolignans of silymarin complex and flavonoid of taxifolin) was evaluated after 6, 12, 24, 48, 72 and 168 hours. Cultures were cultivated on Murashige and Skoog medium with addition 10 mg/ml α-naphtylacetic acid (α- NAA). HPLC method for analysis was used. The maximal content of flavonolignans was reached after elicitor application in concentration c2 after 168 h in callus culture (0,08%). However the highest content of silydianin was detected after application of elicitor in concentration c1 (0.11 %) in suspensions culture. The maximal content of flavonoid taxifolin was noted in of nutritient medium of suspensions cultures.
Possibilities of affecting of secondary metabolites production in culture Silybum marianum in vitro

Lucie Kunzová Possibilities of affecting of secondary metabolites production in culture Silybum marianum in vitro Elicitation is the method making use defensive mechanism of plants to increasing poduction of secondary metabolites in plants and cultures in vitro. The effect of 6, 12, 24, 48, 72 and 168 hours influence by three concentrations of the abiotic elicitor (3-jodo-4-methylfenyl)amide 5-methylpyrazine-2-karboxylic acid on the flavonolignans production in Silybum marianum callus and suspension culture was monitored in this study. The in vitro culture was cultivated on Murashige-Skoog medium with the addition of 10 mg/l of α-naphtylacetic acid as a growth regulator. The content of flavonolignans was determined by HPLC. The maximum content of flavonolignans (0,03 %) in callus culture was demostrated after 24 hours of elicitation (c1 = 2,83 · 10-3 mol/l). The maximum content of flavonolignans (0,04 %) in suspension culture was demonstrated after 48 hours of elicitation (c3 = 2,83 · 10-5 mol/l).
The antioxidant and Hormonal Effects of Ginseng Panax on Lifespan and Stres Resistance in Caenorhabditis elegans

The Thesis: The Antioxidant and Hormonal Effects of Ginseng Panax on Lifespan and Stress Resistance in Caenorhabditis elegans Made by: Václav Ježek The antioxidant and hormonal effect of Panax ginseng on lifespan and stress resistance in C. elegans were investigated. The best results by treatment of C. elegans were achieved in the presence of pure GPE 50 µg/ml without the presence of cholesterol. Surprisingly this medium assigns the increase of mean survival by 10.98 % in contrasting with medium containing the mixture of GPE 50 µg/ml and cholesterol. This negative impact of GPE and cholesterol co- administration on lifespan of C. elegans is more obvious by using sixfold higher concentration of GPE (300 µg/ml). In this case the increase of mean survival by treatment with pure GPE is 17.69 %. In addition the highest increase of progeny was observed in medium with pure GPE 300 µg/ml. Surprisingly this concentration was appointed as toxic with negative effect on lifespan of C. elegans. It is obvious that GPE does not protect C. elegans against oxidative stress significantly because it does not suppress the expression of hsp-16.2/GFP and does not protect against oxidative stress caused by lethal dose of juglone. Name of the work: Effects on flavonoid metabolism and transport in in vitro cultures II. Author: Ludmila Chládeková This diploma thesis is focused on the cultivation of Scutellaria baicalensis Georgii suspension cultures and on the transport of flavonoids through the tonoplast. The effect of MgATP, glutathione and bafilomycine on baicalin and baicalein transport was observed in the isolated vacuoles. Our experiments showed, that MgATP has positive effect on the transport speed and on the total amount of vacuole-stored baicalin. The transport was realized by ATP dependent tonoplast transporters, called ABC transporters. The experiments with glutathione and bafilomycine did not show any effect on flavonoid's transport.

In vitro cultures of medicinal of plants - VIII Method of elicitation is one of the possibilities how to increase production of secondary metabolites in cultures in vitro. The effect of abiotic elicitor (3-jodo-4-methylfenyl)amide 5- terc-butyl-6-chloropyrazine-2-karboxylic acid in three concentrations on the flavonolignans and flavonoid taxifolin production in Silybum marianum callus and suspension culture was tested in this study. Elicitation was run in this time intervals: 6, 12, 24, 48, 72 and 168 hours. Cultivation proceeded on Murashige - Skoog medium with the addition of 10 mg/l of α- naphtylacetic acid. The content of flavonolignans and flavonoid taxifolin was determined by HPLC. The maximum content of flavonolignans (0,05%) in callus culture was found after 168 hours of elicitor treatment in concentration of c2 = 2,33 x 10-4 mol/l. The maximum production of silychristin (0,04%) was found after 6 hours of affecting elicitor c1 = 2,33 x 10-3 mol/l. There wasn't statistical significant increasing of flavonolignans and taxifolin production in suspension culture. The maximum production of silybin A (0,01%) in medium of suspension culture was found after 24 hours of elicitor treatment in concentration of c1 = 2,33 x 10-3 mol/l.
The flavonoid content in commercially available pharmaceuticals containing skullcap

Name of the work: The flavonoid content in commercially available pharmaceuticals containing skullcap Author: Lenka Dvořáková The diploma work was targeted on the determination of flavonoid content in commercially available preparations with content of Scutellaria baicalensis. Two extracts from each preparation were prepared under different conditions. The highest amount of flavonoids was achieved by extraction in ultrasonic bath. Between companies were large differences in measured values as well as among fulfilment of legislative criteria. Dry extracts from China were also tested, because they are used as source material for companies in the Czech Republic. Tested food supplement showed low content of flavonoids and it seems to be poor quality.

My diploma thesis was intended on phytochemical study of extract J1, quaternary acid iodides, of Eschscholzia californica Cham. It concerns the separation of content substances chromatography column with following TLC fraction which was realized at the Department of Pharmaceutical Botany and Ecology. Crystals which were separated from the fraction marked ESCH 23 belong to the group of flavonoids substances. In the concrete, it concerns ferulic acid.
