Substituted oligothiophenes as a building blocks for supramolecular structures preparation

5 Abstract In this thesis the synthesis of (3-bromopropyl) 2,5-bis{5-(2,2':6',2''-terpyridin-4'- yl)-2-thienyl}-3-thienylacetate is reported. As a basic building block it was used 3-thienylacetic acid which is commercially available. The first synthetic sub-step was based on this compound and next synthetic sub-steps included esterification, bromination, borylation or Suzuki coupling. Complexation of synthetized oligomer with zinc cations is also reported, there are included UV-VIS spectra and fluorescent emission spectra of titration range.
Direct arylation and its application in supramolecular polymer synthesis

This thesis is divided into two parts. In the first part there is reported palladium- catalysed regioselective direct arylation of benzofurans with arylsulfonyl chlorides as the coupling partner to C2 carbon. The best reaction conditions of this reaction were investigated, then the influence of various arylsulfonyl chloride substituents to the reaction and finally it is reported direct arylation of C2-arylated benzofuran to C3 carbon to give diarylated benzofuran with different aryl substituents. In the second part, there is reported the synthesis of 2-(hydroxymethyl)-5,7-bis{5- [(2,2':6',2''-terpyridine)-4'-yl]thiophen-2-yl}-2,3-dihydrothieno[3,4-b][1,4]dioxin and modification of its sidechain. As a starting material of the synthesis it was used commercially available 4'-bromo-(2,2':6',2''-terpyridine). The synthetic sub-steps included Suzuki cross- coupling, bromination and C-H bond activation. Then it was also reported the complexation of synthesized oligomer with zinc and iron cations.
Novel hybrid polysaccharide-based polymers for biomedicine

1 Abstract This doctoral thesis is focused on the synthesis and characterization of novel hybrid polysaccharide-based polymers applicable for biomedicine, specifically for a conceptually new bimodal cancer treatment - immunoradiotherapy. For this purpose, polysaccharides β-glucan from Auricularia auricula-judae and κ-carrageenan from Kappaphycus alvarezii, exhibiting immunostimulatory and anticancer activities, were chosen to be grafted with thermoresponsive poly(2-isopropyl-2-oxazoline-co-2-butyl-2-oxazoline)s (POXs) (with different graft lengths and grafting densities) that induced a lower critical solution temperature of the final polymers. The thermoresponsive behavior of resulting polymers was studied with temperature-dependent light scattering methods, fluorescence measurements and also nuclear magnetic spectroscopy to select a polymer material with the most suitable properties for the intended application, aiming at a polymer depot formation after the injection of a polymer solution into the body. The chosen polymer, β-glucan-graft-POX with graft length of 2500 Da, was then modified to bear 1,4,7,10-tetraazacyclododecane-1,4,7,10- tetraacetic acid and a fluorescent dye Dyomics-615 at the graft ends and tested first in vitro to investigate its immunostimulatory properties and also the cellular uptake....
