The role of macrophages in immunosuppression mediated ny regulatory T cells
Kadlecová, Kristýna ; Holáň, Vladimír (advisor) ; Stříž, Ilja (referee)
Regulatory T cells (Treg) represent one of the most important mechanisms of immunoregulation. Treg suppress immune reactions and prevent overactivation of the immune system. There is a lot of ways of Treg action described, here we have focused on Treg interference with macrophages. The suppressor capacity of a highly purified Treg population was demonstrated in proliferation assays. The level of suppression of effector T cell proliferation differs depending on the presence of macrophages in the culture. Treg suppression has been significantly higher in the presence of macrophages. These observations led to hypotesis that Treg affect directly macrophages. However, using flow cytometry, reduction of expression of costimulatory molecules on macrophages after culture with Treg was not observed. Macrophages precultured with Treg showed a comparable functionality as macrophages cultured alone. Neither flow cytometry nor live cell imaging revealed any cytotoxic activity of Treg towards macrophages. Despite the presence of macrophages, Treg did not suppress effector cell proliferation in a model, where stronger activation of effector cells was induced. Therefore, a new hypothesis was presented - initially observed higher suppression in the presence of macrophages was probably caused by a qualitatively or...

Czech healthcare in relation to doctors in hospitals Kristýna Kadlecová Abstract In recent decades, researches focused on doctors attend to the fact that the "unhappy doctors" became a worldwide phenomenon. In the Czech Republic dissatisfaction was publicly revealed in connection with the strike of doctors. The problem that is amplified after transformation are poorly thought out, unilaterally oriented changes in the organizational arrangements that have an impact on hospitals and by extension, as well as a physician. Doctors perform the economic and managerial role in addition to medical one. The key issue is how physicians perceive these pressures and how it affects their working conditions. The aim of this research work is discontent of contemporary Czech doctors working in hospital, i.e. the causes of conflict between their expectations and reality of their labor and other conditions, as developed in the last 20 years, and especially in the last 2 years when there was a radicalization of representation of interests of hospital doctors in the country. In the empirical part is used qualitative research method - semi-structured interviews with physicians. Based on the research, we can observe a situation which devalues the profession of doctor and patient is also affected, which leads into other problems....

Kadlecová, Kristýna ; Pauknerová, Monika (advisor) ; Brodec, Jan (referee)
1 English Summary The aim of this thesis is to compare the determination of law applicable to non- contractual obligations in the Czech Republic and the United States of America. In the beginning I provide a brief overview of the international private law in general and then the substantial law of torts in both Czech Republic and the USA and the prospective changes in the new Civil Code which should be applicapble from 2014. The fifth chapter concerns with the law applicable to non-contractual obligations in the Czech Republic. In the first place there is a statute (Private International Law Act), but the majority of its provisions were overruled by the Rome II regulation, which unifies the privite international law of the EU states. According to the regulation, general rule for the law applicable to non-contractual obligation arising out of torts is lex loci delicti (the law of the country in which the dammage occurs). Rome II then provides special rules for product liability, unfair competition, environmental dammage, etc. In the Czech Republic the regulation does not apply to traffic accidents because the Czech Republic is a contracting state to Hague Convention on the Law Applicable to Traffic Accidents. The sixth chapter focuses on the law applicable to torts in the USA. The first subchapter deals with...

Midwifery care about a woman diagnosed with gestational diabetes mellitus
KADLECOVÁ, Kristýna
Midwifery care about a woman diagnosed with gestational diabetes mellitus Abstract The bachelor thesis is focused on the midwife's care of a woman with a diagnosis of gestational diabetes mellitus. In the theoretical part, there several issues are described, such as gestational diabetes mellitus, its diagnosis and treatment, birth, puerperium, newborn, role of midwife, education, communication and related gestational diabetes mellitus. The survey was conducted with women in their home environment during March 2017. A qualitative method was used to carry out the research part. Data collection was conducted through semi-structured interviews with women diagnosed with gestational diabetes mellitus during pregnancy. Semi-structured interviews were carried out on prior agreement and with the consent of women. The interview contained 8 open questions, which were supplemented, if necessary, with other questions. Data analysis was done by the hand-coded method, or pen and paper method. Answers of individual respondents were identified by codes that were then categorized under each subcategory. One goal was set for this bachelor thesis. The objective was to find out what opinions women have on the possible necessary lifestyle changes in relation to gestational diabetes mellitus. Based on this aim, three research questions have been identified. The first one was focused on how the woman perceived the diagnosis. The second question tried to find out the changes in the lifestyle of women with gestational diabetes mellitus. And the third one was how the woman was educated by a midwife about the lifestyle change. The research revealed that all respondents, after being diagnosed with gestational diabetes mellitus, changed their existing lifestyle. All respondents carefully followed the treatment diet throughout the pregnancy until the end of the puerperium. Of these, two respondents included physical activity in their treatment as a quick walk. A research survey also showed that the midwife gave women limited information about GDM and lifestyle changes. Respondents were totally educated about a lifestyle change at the diabetes clinic, but they did not consider the midwife in the area as a educator. As a result, midwives tend to rely on the education of women from the diabetes clinic, and they only complete women's knowledge. The results of this research can serve ast the information material for midwives who want to improve their care of a woman during pregnancy or who are currently preparing for this profession.
