Cryopreservation Methods in Manufacturing of Advanced Therapy Medicinal Products

This dissertation deals with application of cryopreservation methods in the production and transport of advanced therapy medicinal products (ATMPs), including temporary storage in the Hospital cryostorage facility prior its clinical use. This work kept a strict adherence to European Union regulations and harmonized national legal norms. The first section shows the method validation regarding the collection and cryopreservation of starting material - peripheral blood mononuclear cells for the production of registered ATMPs using chimeric antigen receptor of T-lymphocytes (CAR-T). As a practical output from this endeavor, we obtained the approval for the collection, processing, and distribution of mononuclear cells for ATMP manufacture, as well as an export license to foreign facilities where the final production of genetically modified ATMPs takes place. The second part of this work presents the results of collection of starting material for ATMP production - bone marrow for manufacturing of human mesenchymal stromal cells (hMSCs)-based investigational ATMP and explains the development of the cryopreservation protocol for this investigational product under the conditions of good manufacturing practice (GMP) as an experimental part of the clinical trial EUDRA CT No. 2016-000926-21. Specificity of interrogation of children

62 8. SUMMARY The theme of my Thesis is The Peculiarities of Interrogation of Children. In the introduction of my Thesis I present the reasons which led me to the choice of the theme of my Thesis and describe the division of the Thesis into particular chapters. The second chapter deals with the interrogation in general and its definition, targets, subject and with the division of the interrogation into particular kinds from various points of view. The third chapter contains the interpretation of the term Child in accordance with the Czech law, the criminal procedural regulation of interrogation of children and also the periodization of the child's age based on the stages of development of the human psyche. The forth chapter concerns the preparation of the interrogation, division of the preparation of the interrogation into analytic and synthetic part and with particularities which must be obeyed by the interrogator before the beginning of the interrogation itself. This chapter also deals with the role of the room specially modified for the interrogation of children and the means of the documentation of the interrogation of children. The methods of hospital protection against terrorist attacks by biological weapons

The aim of following work is to pass judgement on how the hospilals are ready to react to possible terrorist attack made with the help of biological materials. Evaluate available crisis management and ability to protect the hospitals and inhabitants from different kinds of terrorist attacks in Czech Republic. The point of this work is at least a bit clear up how biological materials are dangerous and how important is to be at least partly prepared for possible terrorist attack. My work explains term biological terrorism, what is its aim, ahat is the biological weapon, which biological materials can be used as a biological weapon and what kind of effect on humen being is has got. Then my work look on possibilities of terrorist attack and how the hospitals and IZS are prepared for this kind of thread. .
