The influence of simulated body fluids on the apatite production

This bachelor thesis deals with the interaction of various simulated body fluids with ceramic biomaterials. It includes fluid preparation, sample preparation and subsequent analysis of their interaction. Individual samples of biomaterials were exposed to a simulated body fluid for a certain amount of time. After this time, a change in the calcium and phosphorus content of the solutions was determined. At the same time, an electron microscope analysis of sample surface was performed where the dependence of calcium and phosphorus changes was demonstrated. Electron microscopy analysis revealed the formation of new phases on the surface of the studied materials.
Preparation of soil conditioners and characterization of their nutrient release kinetics

This diploma thesis deals with the preparation of multifunctional soil conditioners based on potato starch, in order to slow down the release of nutrients. Materials with different contents of superabsorbent polymer and zeolite were prepared. Spectra indicating the formation of new bonds were evaluated by infrared spectroscopy. The kinetics of nutrient release were observed in the prepared samples of soil conditioners in an aqueous medium, where nutrient retention was demonstrated. Other samples were tested on plants, which proved a mostly positive effect on growth and other factors.
Development of new molecular tools for altering gene expression in Giardia intestinalis .

Giardia intestinalis is a widespread intestinal parasite that causes diarrhea in human and other vertebrate hosts. Although, fully sequenced genome of G. intestinalis has been published, very little is known about the regulation of gene expression. Together with the tetraploid genome, this complicates the use of many common reverse genetics methods. The aim of this thesis was to develop new molecular tools that can be used to alter gene expression in G. intestinalis. For the purposes of this work, new vectors for tetracycline-inducible gene expression including T7 promoter and endogenous oct promoter were designed. Furthermore, cwp1 gene knock-out was created using CRISPR-Cas9 technology. In order to modify mechanisms of double strand break repair, expression of two key components of bacterial NHEJ pathway - LigD and Ku - was introduced into cells of G. intestinalis.

Regulation of gene expression in anaerobic parasitic protist and practical application of knowledge

Because of its importance for every single cell, regulation of gene expression is often studied at all levels today. On the other hand, rather sparse amount of information is available for protists and our understanding of their gene expression is nearly limited to several model organisms. Three anaerobic human parasites have been studied in details: Trichomonas vaginalis, Entamoeba histolytica and Giardia intestinalis. Most recently, some others anaerobic protists (e.g. Entamoeba invadens or some species of Spironucleus genus) have been researched as well. This work is focused on the structure of promoter regions in protein coding genes and application of this knowledge in construction of transfection systems as one of the most effective and powerful tools in molecular and cell biology.
Iron-sulfur proteins and the role of iron in the gene expression of Trichomonas vaginalis

Iron is an essential nutrient for the parasitic protist Trichomonas vaginalis as a component of iron-sulfur (FeS) proteins that are indispensable for energy metabolism of the parasite. The FeS clusters are formed by FeS cluster (ISC) assembly machinery that resides, together with a number of FeS proteins, in Trichomonas hydrogenosomes. These double-membrane bound organelles, which are related to mitochondria, metabolize malate and pyruvate and produce ATP and molecular hydrogen. To obtain more complete information about hydrogenosomal pathways with particular focus on FeS proteins and ISC machinery, we participated on analysis of T. vaginalis genome sequence. To predict hydrogenosomal localization of putative gene products, we developed an application "Hunter" for the in silico searching for N-terminal presequences that are required for protein targeting into the hydrogenosomes. This approach substantially contributed to annotate genes coding for hydrogenosomal proteins that provided base for construction of novel map of hydrogenosomal metabolism as well as for following proteomic studies. Bakalářská práce Systém sociálních služeb poskytovaných v oblasti péče o uprchlíky v České republice se věnuje tématu systému sociálních služeb poskytovaných v oblasti péče o uprchlíky v České republice. Jeho popisu, kam dospěl jeho patnáctiletý vývoj, co jsou jeho subjekty a jak spolupracují. Zkoumá také otázku zda vede tato péče k závislosti na sociálních službách, zda má negativní dopad na své klienty. Pokud ano, v čem toto úskalí spočívá - spíše v systémové chybě nebo špatném přístupu jednotlivých pracovníků. Obecná část práce je zaměřena na nastínění základních charakteristik dané oblasti, především na její specifika a jaký to má vliv na práci s klienty. Dále se věnuje několika současným trendům objevujícím se v oblasti migrace a péče o uprchlíky v České republice. Konkrétně migračním politice, postoji veřejnosti k cizincům, finanční prostředkům poskytovaným na péči o uprchlíky a vývoji počtu uprchlíků. Empirická část navazuje zkoumáním vyvstalých otázek z části obecné a to na postojích pracovníků nevládních neziskových organizací. Lze shrnout, že pracovníci vnímají negativní dopad systému a péče na uprchlíky. Příčiny vidí ve špatně definované migrační politice, nedostatečné komunikaci mezi jednotlivými subjekty, v jejich nerovnoprávném postavení, špatném přerozdělování finančních prostředků,...
