Biotechnological production of PHA employing Cupriavidus malaysiensis

The subject of this diploma thesis is the study of the production of polyhydroxyalkanoates (PHA) employing Cupriavidus malaysiensis. The aim of this thesis is to obtain and characterize the polymer with the most advantageous properties. The theoretical part deals with polyhydroxyalkanoates and their biotechnological production. Then selected microorganisms producing 3-hydroxybutyrate and 4 hydroxybutyrate, P(3HB co 4HB), are described. After that, the theoretical part deals with the production of PHA employing Cupriavidus malaysiensis. The bacterial strains of Cupriavidus malaysiensis DSM 19416, 19379 and 25816 were used in the experimental part. The strain Cupriavidus necator H16 was used as a control strain. First, the ability to utilize various carbon sources by the given bacterial strains was determined. Then, the aim was to select the optimal carbon source, a precursor for the synthesis of a polymer with high content of 4-hydroxybutyrate, and to determine the ideal culturing conditions for the growth of the microorganisms, which have been studied, and the production of P(3HB co-4HB). A two-stage culture and bioreactor culture was also performed to produce the desired copolymer. Furthermore, the production of terpolymer of 3-hydroxybutyrate, 4 hydroxybutyrate and 3-hydroxyvalerate, P(3HB co 4HB co 3HV), was studied by single stage and two-stage cultivation. In the last part, the produced polymers were characterized by gas chromatography with flame ionization detector (GC-FID), size exclusion chromatography with multi-angle static light scattering (SEC-MALS), differential scanning calorimetry (DSC) and thermogravimetry.
Biopolymers produced by halophillic bacteria and their role in stress response

The subject of this thesis is the study of halophilic microorganisms. The main part deals with isolation and characterizatin of the extracellular polysaccharide (EPS) produced by the archeal strain Haloferax mediteranei. Chemical structure was determined by Raman spectroscopy, FTIR and elementar analysis. Very low amount of sulphate groups indicate, that generally accepted structure of the EPS is not universal and can be dependent upon cultivation condition. Further, EPS revealed substantial protective effect for both model prokaryotic organisms – Cupriavidus necator and model eukarytoic microbe – Saccharomyces cerevisiae when the cells were exposed to various stress factors such as freezing, high temperature, osmotic pressure or ethanol.

The focus of master thesis are hedgerows in the landscape. The aim is to describe particular aspects of hedgerows in the landscape and define their structure and spatial arrangement. The main output is project documentation containing draft hedgerow and reflecting the theoretical findings. Thesis should also contribute to enhancing the public awareness of this important element of scattered vegetation.

Importance of Identity in Nursing Profession and its Impact on Quality of Care

The nursing profession belongs to helping professions which are heavily demanding in terms of mental and physical aspects. The nurses who are university graduates enter into practice more mentally and physically mature, and their approach to patients and the care provided may be better than in nurses with secondary education. In care provision nurses have to cooperate with physicians, but also with other healthcare professionals. Due to the lack of medical personnel nurses assume responsibilities of other team members, which they may perceive as a restriction in fulfilling their professional identity. This restriction may be reflected in the quality of the care provided. It often happens that a nurse cannot fulfill her professional identity because of the pressure of her family which requires the fulfillment of their own requirements prior to fulfilling the professional identity of a nurse.The aim of the thesis was to determine how nurses, but also their family members, perceive the professional identity of nurses. The aim was achieved. The research was carried out in two phases. The first phase was conducted using interviews with six nurses of different ages both with and without university education. The second phase was carried out through a quantitative survey using a questionnaire method in health care facilities in Písek, České Budějovice and Plzeň. Results of the research can be used in qualification and lifelong education of nurses to strengthen their identification with the profession.
Problems of nursing care about patient with apalic syndrom

This thesis deals with problems in nursing care of clients with apallic syndrome. There were 4 hypotheses. The first hypothesis presumes that nurses give patients care focused mostly on treating developed complications. After evaluation of the results focused on nursing care, we found out that some nurses know the correct processes while others do not. This means that patients are sometimes given the correct care and sometimes not. So hypothesis 1 was proved valid. The presumption for the other three hypotheses was that nurses do not have information about BS conception, do not know about possibilities of its usage, and do not use this conception in their care of clients with AS. However, the results concerning BS conception were very positive. Nurses know the conception and use it in their care about clients with AS. Hypotheses 2, 3, and 4 were not proved valid.
