DOES ONE MONTH CREATINE SUPPLEMENTATION IMPROVE BLOOD LIPIDS?
Kohlíková, E. ; Petr, M. ; Navrátil, Tomáš ; Heyrovský, Michael ; Přistoupilová, K. ; Přistoupil, T. I.
One month’s administration of creatine (CR) in the dose of 5 g/day to 11 young active sportsmen affected their daily amount of CR excreted into urine, and the levels of cholesterol (CH), of triacylglycerols (TAG), of vitamin B12 and of folates in blood. The probands were divided into 4 groups, according to the amount of CR found in urine, and of folates and vitamin B12 determined in blood. It is possible that added CR might induce individually higher offer of 2C units, released under catalysis of vitamin B12 from HoCySH and serine in enzymatic reaction catalyzed by CBS, and in this way higher offer of cysteine and succinyl-CoA This is the main effect of CR in the group D. Both vitamins influence the ratio between glycine (necessary mainly for CR synthesis, beside other metabolic pathways) and serine. The change of serine into glycine can be realized by THF, under catalysis of serine hydroxymethyltransferase. For the opposite change of glycine into serine the metabolic pathway is catalyzed by folates too, but in more complicated way
Suplementace kreatinu u sportovce s hyperhomocysteinemií

One month's administration of creatine (CR) in the dose of 5 g/day to 11 young active sportsmen affected their daily amount of CR and creatinine excreted into urine. The probands were divided into 4 groups, according to the amount of CR found in urine, and of folates and of vitamin B12 determined in blood. The ganges of folates and vitamin B12 were mutually reciprocal. Each group utilized CR as donor of one- and two-carbon (1C and 2C) units by means of homocysteine (HoCySH), folates, and vitamin B12, in different metabolic pathways. In 10 men the CR administration was accompanied by an increase of HoCySH level in blood, while for the last man, with accidentally discovered hyperhomocysteinemia, after the month's CR administration the HoCySH level dropped by 50 %.

Voltametrické stanovení karcinogenních vzduného polutantu 2-nitrofluorenu za použití leštěné stříbrné pevné amalgámové kompozitní elektrody
Vyskočil, V. ; Krejčová, Z. ; Daňhel, A. ; Navrátil, Tomáš ; Barek, J.
The emissions of gasoline and diesel engines contribute significantly to ever increasing pollution of living environment. Studied compound - 2 nitrofluorene (2 NF) - is proven to be carcinogenic to rats and thus possibly carcinogenic to humans. The procedure of determination of 2-NF using polished silver solid amalgam composite electrode (p AgSA-CE) was developed.
Význam lipidů v metabolismu tuků
Kohlíková, E. ; Petr, M. ; Přistoupilová, K. ; Přistoupil, T. I. ; Pelclová, D. ; Žák, J. ; Heyrovský, Michael ; Navrátil, Tomáš
In course of last two years we compared values of 34 parameters, obtained from 11 young active sportsmen given 5 g of creatine (CR) daily during 1 month. Among these parameters were the products of metabolic pathways, the changes of which are connected with the development of atherosclerosis. This holds for the levels of cholesterol (CH), triacylglycerols (TAG) and homocysteine (HoCySH), and for the levels of vitamin B12 and of folates in blood.
Preparation and Properties of Mercury Films on Solid Amalgam Surface
Josypčuk, Bohdan
There have been suggested a simple electrolyzer and a reliable procedure for preparation of mercury films with precisely defined thickness on a conducting surface. A small volume of electrolyte which can be used more times and a paste amalgam which cannot spill substantially decrease a risk of environment contamination. Measurements were parallelly performed on a liquid mercury free silver solid amalgam electrode (AgSAE), on a mercury meniscus modified AgSAE and on HMDE for comparing their properties with those of the mercury film electrodes (MF-AgSAE). A current response of the film electrodes is the biggest. The peak width, which determines the method resolution, is smallest on the film electrodes. Range of working potentials on MF-AgSAE is comparable with one on HMDE in different supporting electrolytes.
The effect of working electrode on electrolytic reaction
Josypčuk, Bohdan ; Heyrovský, Michael
The courses of a) redox reaction of ferrocene, b) redox reaction of thallium ion, c) electroly- tic reduction of 4-nitrophenol, were followed with 14 different working electrodes. The effect of the working electrode on electrolytic reaction is in each case specific, sometimes it is negligible, but it should be always considered.
Patch Clamp Technique – The Tool for Ions Transport across Model and Real Biological Membranes
Navrátil, Tomáš ; Jaklová Dytrtová, J. ; Jakl, M. ; Šestáková, Ivana ; Mareček, Vladimír ; Krůšek, Jan
The project is aimed at the preparation and characterization of model and/or real phospholipid bilayers at stabilized phase boundaries enabling studies of transfer of charged and uncharged species between the two phases. The best way for investigation of inorganic ion transfer is provided by using mercury as one of the phases, in view of the large amount of the relevant electrochemical data available, whereas the transfer of more complex species is better studied in the system of an organic and an aqueous phase stabilized by (agar) gel. The results contribute to the understanding of ion transport across real membranes. Polypeptides and peptides capable forming ionic channels and enabling transport of various ions and molecules across the membranes, will be incorporated into the model membranes. The possibilities of transport of selected compounds across the model membrane will be investigated using cell permeable peptides as well.
The Use of Electrochemical Stripping Methods for Lead and Cadmium Speciation in Soil Solutions
Šestáková, Ivana ; Jaklová Dytrtová, J. ; Jakl, M. ; Navrátil, Tomáš ; Fojta, Miroslav
Soil solution is an aqueous phase of soil which is affected by plant root systems. The soil solution contains nutrients, organic and inorganic soil substances and also exudates – substances released by plant roots (e.g. low molecular weight organic acids). For the bioaccumulation (transport from soil into plants) the free metal ions and labile complexes contents (usually in trace amounts) in soil solution are important. That is why the electrochemical methods using working electrode preconcentration step, i.e. differential pulse anodic stripping voltammetry, anodic stripping square-wave voltammetry and stripping chronopotentiometry with constant current, are applicable. Mentioned methods combined with the hanging mercury drop electrode were tested for the cadmium and lead speciation possibilities in model and also real soil solutions.
Electrochemical detection of trace amounts of nitrated explosives
Barek, J. ; Fischer, J. ; Vyskočil, V. ; Pecková, K. ; Navrátil, Tomáš ; Josypčuk, Bohdan
Nitrated organic compounds belong to the group of the most frequently used explosives. Detection of low levels of such compounds is very attractive in these days. Due to the easy reducibility of the nitro group, voltammetry seems to be very suitable method for these purposes. For these determinations the following electrodes were used: were used hanging mercury drop electrode, meniscus modified silver solid amalgam electrode, miniaturized paste silver amalgam electrode, paste electrode on the basis of silver amalgam powder and proper organic liquid were used.
