Assessment of contamination around the landfill Pozďátky

This bachelor thesis is focused on the characterization of the locality and on the evaluation of water contamination in a short distance from Pozďátky - the hazardous landfill. The occurrence of the selected contaminants is assessed. The samples were collected at five different locations: landfill leaks, the confluence of these leaks with the Prašinec creek, in other parts of the Prašinec creek affected by contaminated waters and on the very site of the creek Prašinec where no water pollution has been expected.

Voltage sensitive fluorescent protein based on the motor protein prestin

The aim of this work was to create a series of fluorescently labeled constructs based on the motor protein prestin, express these constructs in mammalian cells, and measure their linear dichroism by two photon polarization microscopy, in order to develop a genetically encoded optical probe of cell membrane voltage. This thesis provides an introduction to current methods of observing electrical activity in neurons, describes the method of two-photon polarization microscopy, describes our results in preparing the abovementioned constructs, and summarizes our microscopic observations.

Tax expenditures as an instrument of tax policy

The work deals with an aspect of the tax system, the tax expenditures. The aim of the work is to estimate selected tax expenditures that are typical for the personal income tax in the Czech Republic, and to devise methods of its calculation. The introductory chapter deals with the definition of the tax expenditure, its types, methods of its calculation, and the effects of tax expenditures on the national budget. The second part of the work comprises the analysis of tax expenditures related to the personal income tax. The summary of the analysis and comparison of tax expenditures with Australia and Canada is presented in the third part, and the main ideas of the upcoming tax reform are summarized in the final part of the work.
