Possibilities of hematopoietic stem cells determination in the peripheral bood.

In this study the hematopoietic stem cell, its properties, sources, utilization and possibilities of its determination in the peripheral blood are described. The detection of CD34+ cells by flow cytometry is the reference method for determining the number of hematopoietic stem cells. The aim of my work was to verify whether it is possible to identify such cells can also use another method. This method is the determination of hematopoietic progenitor cells by a haematology analyser Sysmex XE 2100. In the practical part of the study, data of patients obtained from the analyzer Sysmex XE 2100 and flow cytometer are processed. This is the number of hematopoietic progenitor cells, CD34+ cells and total number of leukocytes. The data are compared by using the Spearman correlation test. It has been shown that the numbers of hematopoietic progenitor cells and CD34+ cell counts, there is dependence. Correlation coefficients of 0.55 and 0.58 can be considered very well. The analyzer Sysmex XE 2100 can be used as an alternative method to determine the number of hematopoietic stem cells in peripheral blood and leukoconcentrate.
