Design of Power HIL unit for Li-pol cell simulation

This thesis deals with design and construction of a Li-po cell Power-HIL simulator intended for testing of a small wireless module's powering. The simulator is based on a battery model which includes the influence of temperature and battery degradation on battery parameters. To estimate model parameters a series of tests on real Li-po cell with capacity 250mAh were performed. Charging and discharging tests were performed by different temperatures of the battery. In purpose of this test a simple thermal chamber was constructed. Design of the power electronics of the device was tested in the Simscape library of the Matlab Simulink. The result is a working prototype of the simulator tested for charging and discharging of batteries with capacity in range 100-1500 mAh. The simulator can be controlled with graphical user interface capable of setting the battery parameters and real time observation of the model variables.
Influence of cultivation conditions on the production of recombinant proteins

Recombinant proteins are produced by using genetic modifications. In this process is insert contained gene encoding a certain protein in a cloning vector cloned into the host organism. Recombinant proteins are expressed after the transformation of the cloning vector into a host, the host organism. The expression of recombinant proteins in bacterial cells is one of the most efficient ways to manufacture these proteins. The p53 protein is a tumor suppressor protein, the main role in cells is to react on DNA damage. Due to the reaction to various intracellular and extracellular stimuli, including DNA damage, the p53 protein shows different biological functions, including regulation of senescence, cell cycle, or apoptosis. The theoretical part of the thesis part presents the basic properties of proteins, methods of recombinant protein expression, methods of protein isolation, and characterization of p53 protein. The aim of the experimental part was to determine the effect of incubation temperature on recombinant p53 protein production. The work involves the isolation of plasmid DNA and its transformation into E. coli production cells. The produced proteins were successfully isolated and subsequently characterized by SDS-PAGE and Western Blotting.
Maziva vhodná pro tažení oceli

Cílem práce je experimentální vyhodnocení vhodnosti maziv pro tažení korozivzdorné oceli 1.4301, a to za zvýšené teploty, která odpovídá teplotě v reálném procesu. Metoda testování maziv je založena na modelování tření mezi tažnou hranou a přidržovačem, které odpovídá reálnému procesu. Konstrukce zkušebního přípravku proto také odpovídala tomuto modelu. Výsledkem práce je porovnání vhodnosti jednotlivých maziv s referenčním mazivem, používaným ve společnosti ACO Industries. Nejlepší výsledky byly naměřeny u maziva AT Kalorex 650 S, které obsahuje ekologicky závadné přísady.
