System for maintaining the sharpness of the image for the light microscope

Live imaging is a key tool for modern biological discoveries. However, without techniques for maintaining the sample in focus is hardly feasible. In this thesis I present an application of the optical vortices for the sample axial localization, so it will be maintained in focus. Experimental setup from previous experiments has been redesigned, so it could be integrated into the microscope. The setup has been built and successfully tested. To do this, the control software also had to be written. Also, feedback script for future integration into the microscope has been prepared.
