A pathway during transient differentiation of human colon cancer HT-29 cells is signallized by an activation of the protein kinase B and glycogen synthase kinase-3. Their phosphoacceptor sites are reproduced with synthetic peptides.

Using synthetic peptides which reproduce phosphoacceptor sites in the molecule of GSK-3, PKB an eIF2B, we found that phosphorylation and activity of PKB increased very early during in vitro differentiation induced by sodium butyrate. This was inhibited by wortmannin, the selective inhibitor of phosphoinositol-3 kinase, indicating that the control of PI3 kinase/PKB/GSK-3 signalling pathway is the early event of transient differentiation of the human colon cancer cells HT-29.
