Separation of tryptic digest of cytochrome c within a long nanoelectrospray tip

Capillary electrophoresis coupled to mass spectrometry (CE-MS) posses powerful tool for separation and detection of wide range of ionic species. To avoid complicated constructions of interfaces we have conducted the separation within a long and thin nanoelectrospray tip [1]. We have investigated main properties of such an experimental design on a complex sample of tryptic digest of cytochrome c.
Analysis of saccharides in grapes and wine using HPLC with various stationary phases

The quality of wine is influenced by its saccharides content, especially glucose and fructose. Monitoring of wine oligosaccharides is of great interest, because of their physiological, medical and food importance. The aim of study was to develop method for fast analysis of saccharide composition of wine, grape juice and grapes.

Development of sensitive, miniaturized and fast methods aims to reliable analyze even small amounts of samples. Micromanipulation systems such as laser capture microdissection enable to collect just single cells from cell suspensions or tissue samples. One option for further analysis is the bioluminescence reaction based on luciferin/luciferase conversion. This approach was used for detection of active caspases, caspase-3/7. Droplets based microfluidic systems provide benefits in biological, pharmaceutical and chemical research.
Subcritical extraction as sample treatment method for antioxidant screening of various plant extracts

Pressurized Hot Water Extraction performed in both static and dynamic modes followed by HPLC with UV-ViS detection was employed for fast and efficient determination of rutin and quercetin from various berries. Moreover, the antioxidant power of subcritical extracts applying •DPPH was evaluated.
ZIC-HILIC monolithic capillary column coupled with MALDI-MS: A tool for glycan analysis

In this contribution, we report analysis of glycans enzymatically released from bovine ribonuclease B (RNase B) and human immunoglobulin G (hIgG) combining glycan separation using the synthesized zwitterionic silica-based monolithic capillary column and off-line MALDI-MS detection.

Low-cost 3D-printed fluorescence detector for capillary electrophoresis

We have verified the possibility of Fused Deposition Modeling (type of additive manufacturing) for lab-made construction of inexpensive fluorescence detector for capillary electrophoresis using commercially available optical components. Sensitivity of fabricated and assembled detection system was tested by fluorescein solutions. Limits of detection were determined for several tested arrangements.
Polydopamine coated capillaries for CE separations

We have tested CE separation of selected samples in capillaries with polydopamine modified surface. The capillaries were modified by polydopamine or polydopamine with an additive. The polydopamine coating with additive represents a simple and effective procedure for capillary alteration by another modificator such as hydroxyethyl cellulose, hydroxypropyl cellulose etc. In this work, we represent separation data from PrinCE system with UV detection for peptides, proteins and oligosaccharides labeled by 2-aminobenzoic acid.
IEF and HPLC-based methods for efficient bacterial characterization

Pectobacterium and Dickeya species are responsible for blackleg and tuber soft rot diseases on crop and ornamental plants. Recently, a more virulent and aggressive Dickeya type of E. chrysanthemi was described and it becomes serious problem for potato production in Europe. In this study, we suggest procedures using two commonly available laboratory techniques, gel isoelectric focusing and liquid chromatography, for differentiation and characterization of Pectobacterium and Dickeya species. We have confirmed that fingerprinting approach can be used for bacterial differentiation, which can be further used in rapid diagnosis of plant disease.
Porous layer open tubular columns with immobilized trypsin for protein digestion

We have developed a monolithic porous layer open tubular (PLOT) column with immobilized trypsin for protein digestion. The PLOT column was prepared in a 10 (mikro)m ID fused silica capillary. Trypsin was immobilized on the monolithic surface and the developed enzyme reactor was used for protein digestion followed by on-line ESI/MS analysis.
