Detekce prionových proteinů a jejich interakce s kovy a metalothioneinem

Prion diseases are formed by a conformational change of prion-like protein (PrPC) with alfa-helix structure to the pathological isoform - prion (PrPSc) which acquires beta-sheet structure. PrPC physiological properties in the brain are insufficiently described but there is an assumption of its affinity to metal ions. Another protein with metal-binding ability is metallothionein (MT). Brain specific isoform of MT is called MT-III and it is assumed to participate in maintenance of metal ions concentration in the brain. Aim of this study was to prepare recombinant human PrPC in E. coli. Furthermore, this protein was used to detect interactions between metal ions (Cu, Zn), MT and PrPC by differential pulse voltammetry method. The final part was devoted to the MT-III determination in different genotypes of prion-infected and non-infectious mouse brain tissues.

The thesis deals with current opportunities for investing in precious and industrial metals. The literature overview introduces the reader with the current knowledge about commodity investments and provides results of various authors dealing with factors important for commodity price develop and with relation of commodity prices and inflation. Trough metal markets analysis identifies key factors with impact on sup-ply, demand and price of specific metal. Based on historical prices explores the relationship of metal prices, relationship of metal prices and stock market and business cycle. Quantify risk, which the investor has to accept in the case of investing in metals by using Value at Risk methods. The risk is compared with the level of risk on stock market. The thesis summarises all these results and formulate investment recommendation.
Long-term development of raw materials prices

This bachelor thesis focuses on long-term development of raw materials and connected with them Simon - Ehrlich bet. Basic data were obtained from accessible databases USGS. The aim of this thesis is by statistical methods to survey the development of prices five metals: copper, tungsten, tin,chromium and nickel, compare the data with an average income and money supply and try to extend famous Simon-Ehrlich wager to 2010 year. This thesis is divided into five parts. In the first two is described wager a background information of the development of metals and their prices. The statistical methods used in the work and their use to equalize basic data are described in the third and fourth chapters. The fifth chapter is an overview of prolongation bet in 1990-2010 period.
